Design, synthesis and antibacterial evaluation of a polycationic calix[4]arene derivative alone and in combination with antibiotics

Abstract: The growing antibiotic resistance phenomenon continues to stimulate the search for new compounds and strategies to combat bacterial infections. In this study, we designed and synthesized a new polycationic macrocyclic compound () bearing four -methyldiethanol ammonium groups clustered and circularly organized by a calix[4]arene scaffold. The activity of compound , alone and in combination with known antibiotics (ofloxacin, chloramphenicol or tetracycline), was assessed against strains of (ATCC 6538 and methici… Show more

“…To investigate the antibacterial activity of the MgONPs and bulk MgO, these materials were diluted in a series of concentrations ranging from 15.625 to 1000 μg/mL (1000, 500, 250, 125, 62.5, 31.25, and 15.625 μg/mL), and a typical microdilution method was performed to determine the MIC and MBC of these chemicals against R. solanacearum ( Consoli et al, 2018 ). The MIC and MBC were determined to indicate the bacteriostatic and bactericidal activity.…”

“…Furthermore, the antibacterial activity of MgONFs against Xoo strain GZ 0005 was determined in 96-well microtiter plates by measuring the OD 600 using a microplate reader, which was carried out as described by Consoli et al [25]. In brief, each well of 96-well microtiter plates was filled with 100 μ l solution of MgONFs, which were suspended in NB broth to give a final concentration of 4, 8, and 16 μ g/ml.…”

“…An ideal antimicrobial will display specific activity against one species of bacteria, while having no effects on any other organisms, whether they be humans being treated for an infection or the diversity of animal and plant life that may exist in a field where crops are sprayed. An encouraging study into the toxicity and biodistribution of p-sulfonatocalix [4]arene in mice by Coleman and colleagues using a 34 S-labelled derivative found no toxic effects in mice up to 100 mg kg −1 of body weight ( Figure 2) [12]. One animal was given a dose of 400 mg kg −1 and died within an hour, whereas 14 others, receiving doses of 40 or 100 mg kg −1 , were outwardly unaffected.…”

“…A polycationic calix [4]arene derivative, 46 ( Figure 9), was prepared by Nostro and colleagues, through the reaction of N-methyldiethanolamine on tetra-propoxy-4-chloromethylcalix [4]arene, to deliver ofloxacin, chloramphenicol, and tetracycline [34]. The calixarene alone had a similar profile to chloramphenicol against S. aureus ATCC 6538, but its MIC was lower by a factor of two against MRSA 15, MRSE 17, and P. aeruginosa isolate 1.…”
