Design and synthesis of Raman reporter molecules for tissue imaging by immuno‐SERS microscopy

Abstract: The design and synthesis of Raman reporter molecules comprising olefin or alkyne moieties with strong and characteristic vibrational Raman bands is presented. Chemisorption onto the surface of colloidal Au/Ag shells yields a self-assembled monolayer. Hydrophilic stabilization of such SERS labels can be achieved by short terminal ethylene glycol units attached to the Raman reporter. Encapsulation by silica with subsequent functionalization of the glass surface allows the conjugation to biomolecules such as anti… Show more

“…Minimal sampling was suggested for rapid acquisitions of Raman images [104]. Raman signals can be enhanced in tissue or cells by surface enhanced Raman scattering (SERS) effect using functionalized metal nanoparticles [105][106][107] or by resonance Raman scattering probing prosthetic groups in chromophores [108]. Another promising signal enhancement approach constitutes non-linear, coherent Raman techniques such as coherent anti-Stokes Raman scattering (CARS) or stimulated Raman scattering [109].…”

“…A stoichiometric ratio of Ra 1 : Ra 2 ¼ 100 : 1 was chosen to ensure the steric accessibility of the carboxyl groups in the densely packed SAM. 19,20,39 The addition of DTNB-MEG-OH (Ra 1 ) and DTNB-TEG-CO 2 H (Ra 2 ) to the metal colloid leads to a hydrophilically stabilized self-assembled monolayer comprising two different terminal spacer units (dual spacer-SAM, cf. “…The approach may also lack spectral resolution in some applications where characteristic reporter bands (such as the phenyl ring modes of 4-MBA) only exhibit small wavenumber shifts (few cm –1 ) as a function of bound substituents. 155 158 …”
