While the general effect of CO2 enrichment on photosynthesis, stomatal conductance, nitrogen elemental content or yield has been documented, there is still some uncertainty as to whether there are interactive effects between CO2 enrichment and other factors (such as temperature, geographical location, water availability and cultivar). In addition, the metabolic coordination between leaves and grains, which is crucial for crop responsiveness to elevated CO2, has never been examined closely. Here, we addressed these two aspects by taking advantage of several FACE experiments across five different countries using multi-level analyses. There was little effect of elevated CO2 on yield (except in USA) likely due to photosynthetic capacity acclimation, as reflected by protein profiles. Also, there was a significant decrease in leaf amino acids (threonine) and macro-elements (such as K) at elevated CO2, while other elements such as magnesium (Mg) or sulfur (S) increased. Despite the insignificant effect of CO2-enrichment on yield, grains appeared to be significantly depleted (as expected) in N, but also in threonine, the S-containing amino acid methionine and Mg. Overall, our results suggest a strong detrimental effect of CO2 enrichment on nutrient availability and remobilization from leaves to grains.

Increasing atmospheric CO and drought are major symptoms of anthropogenic climate change with profound effects on plant growth. Transgenerational memory (i.e. influence of the parental environment on offspring phenotype and performance) has been suggested as a relevant mechanism for plants to build-up adaptative capacity for rapid environmental changes. However, this mechanism of pre-adaptation remains poorly investigated so far. We investigated intra- and transgenerational effects of elevated CO on drought response of wheat. We used seeds from a FACE (Free Air Carbon Dioxide Enrichment) experiment with ambient and elevated CO to grow plants in climate chambers in which we varied CO , atmospheric water demand and soil moisture. We quantified photosynthetic efficiency, stomatal sensitivity and biomass production. We observed intragenerational upregulation of photosynthetic efficiency but transgenerational downregulation of photosynthetic efficiency, stomatal sensitivity and water use efficiency as response to maternally elevated CO . Plant biomass was affected by drought and experimental CO but not by maternal CO . Our study showcases the importance of transgenerational memory effects when studying climate change response of plants and could have major implications for our understanding of global dynamics of carbon sequestration. It highlights the pressing need for multi-generational experiments accounting for transgenerational memory effects of elevated CO .

A simple cloud point preconcentration method was developed and validated for the determination of gallic acid, bergenin, quercitrin, and embelin in Ardisia japonica by high-performance liquid chromatography (HPLC) using ultrasonic assisted micellar extraction. Nonionic surfactant Genapol X-080 was selected as the extraction solvent. The effects of various experimental conditions such as the type and concentration of surfactant and salt, temperature, and solution pH on the extraction of these components were studied to optimize the conditions of Ardisia japonica. The solution was incubated in a thermostatic water bath at 60°C for 10 min, and 35% NaH2PO4 (w/v) was added to the solution to promote the phase separation and increase the preconcentration factor. The intraday and interday precision (RSD) were both below 5.0% and the limits of detection (LOD) for the analytes were between 10 and 20 ng·mL−1. The proposed method provides a simple, efficient, and organic solvent-free method to analyze gallic acid, bergenin, quercitrin, and embelin for the quality control of Ardisia japonica.
