Suspensions are two phase systems composed of solid materials dispersed in liquid. Detarium microcarpum gum (DMG) was obtained by acetone precipitation of filtrate obtained from maceration of Detarium microcarpum seed powder using distilled water. Ibuprofen was pulverized using a mortar and dispersed in distilled water to form ibuprofen suspension formulation (IS1). Five other ibuprofen suspension formulations were prepared using either DMG or acacia as the suspending agent (IS2-IS6). Benzoic acid was added during the preparation of formulations IS4 and IS6 as preservative. Physicochemical properties like pH, viscosity and the total microbial count for the suspensions were evaluated. Drug-excipient compatibility was studied using FTIR spectroscopy. The pH of the suspensions ranged from 3.7±0.0 -5.3±0.0 on day 0 to 3.9±0.0 - 6.3±0.0 on day 35. Flow rate was from 2.21±0.13 – 33.00±3.70 mls-1 on day 0 to 1.88±0.12 – 20.19±0.16 mls-1 on day 35. The viscosity at 25oC ranged from 2391.3 – 5702.2 mPas on day 0 to 3793.0 – 3828.2 mPas on day 35. It was a shear thinning system. Formulation IS6 was the most stable and IS1 was the least stable. The suspensions contained high level of bacteria on day 0 which increased significantly on day 35, with exception of formulations IS4 and IS6. Ibuprofen suspensions with good physicochemical properties were produced using DMG but the level of microbial contamination observed highlighted the need of having good environmental hygiene and use of preservatives when natural gums are used in formulations. This research was done to formulate and assess the sustained release property of metformin tablets prepared from metformin microcapsules produced by ionic gelation technique using Sida acuta gum or sodium carboxymethylcellulose as release retardant. Sida acuta gum was produced by isopropyl alcohol precipitation of the filtrate obtained from distilled water maceration of powdered dried Sida acuta leaves. Metformin and Sida acuta gum compatibility was determined using FTIR. Metformin microcapsules were prepared by ionic gelation technique using sodium alginate alone or with either Sida acuta gum or sodium carboxymethylcellulose as release retardant. The microcapsules were evaluated for % yield and flow properties. The size and surface morphology of the microcapsules were determined using scanning electron microscopy. Metformin tablets were prepared from the microcapsules using direct compression technique. The tablets were evaluated for hardness, friability and in vitro dissolution. There was no major incompatibility between metformin and Sida acuta gum. The microcapsules have excellent flow property and were of micrometer size range. They have rough surfaces. The friability of the tablets ranged from 0.4-1.6% while the hardness was from 3.82-11.17 Kgf. The tablets from formulations M2 failed both friability and hardness test. About 44.5-47.1% of metformin was released from the tablet formulations after 6 h while 86.0-100% was released after 10 h. All the tablet formulations showed sustained release ability. To improve the compliance of patients with type 2 diabetes there is a need to develop metformin oral dissolving tablets, which disintegrate and dissolve rapidly in the saliva. The aim of this study was to optimize the composition of metformin orodispersible tablets. The central composite design was used to establish the relation between independent variables, such as, quantity of MCC Burst, quantity of Neusillin US2, and dependent variables, such as, flowability, bulk density, tapped density, uniformity of weight, friability, tablet hardness, disintegration time, wetting time in order to obtain the optimal formulation using Response Surface Methodology. After generating the polynomial equations that relate the dependent and independent variables, the process was optimized for five responses. From the dissolution studies, it is confirmed that all formulations released more than 82% of the metformin within 3 min. It was found that the tablet containing 3% MCC Burst and 5% Neusillin US2 was a better formulation in terms of hardness (72 N), uniformity of weight (1.09%), friability (0.75%), rapid disintegration (33 sec) and drug release (95.0%) when compared with all other formulations. RezumatPentru a îmbunătăți complianța pacienților cu diabet zaharat de tip 2, este nevoie de a dezvolta comprimate cu metformin cu dizolvare orală, care se dezintegrează și se dizolvă rapid în salivă. Scopul acestui studiu a fost de a optimiza compoziția comprimatelor orodispersabile cu metformin. Am stabilit relația dintre variabile independente, cum ar fi, cantitatea de MCC Burst, cantitatea de Neusillin US2 și variabilele dependente, cum ar fi curgerea, densitatea în vrac, densitatea la tasare, uniformitatea masei, friabilitatea, rezistența la rupere a comprimatelor, timpul de dezintegrare, timpul de umectare pentru a obține formularea optimă folosind metodologia suprafeței de răspuns. După generarea ecuațiilor polinomiale care leagă variabilele dependente și independente, procesul a fost optimizat pentru cinci răspunsuri. Studiile de dizolvare confirmă că toate formulările au eliberat mai mult de 82% din metformin în 3 minute. S-a constatat că tabletele care conțin 3% MCC Burst și 5% Neusillin US2 au prezentat o formulare mai bună în ceea ce privește duritatea (72 N), uniformitatea masei (1,09%), friabilitatea (0,75%), dezintegrarea rapidă (33 sec) și eliberarea substanței active (95,0%) în comparație cu toate celelalte formulări evaluate.
