Design and development of tofacitinib citrate loaded transferosomal gel for skin cancer by box-Behnken design- doe approach

Abstract: Objective: The purpose of the present study is to develop, statistically optimize and evaluate the transfersomal gel formulation of Tofacitinib citrate via transdermal route by DOE Approach. Materials and Methods: Transferosomes are made up of with bilayer forming phospholipids and biocompatible surfactants like sodium deoxycholate. The surfactants also acts as edge activator which stabilizes the phospholipid bilayer and increases the deformability of the vesicle. The formulation were designed by Box-Behnken D… Show more

“…As represented in Table 7 , the spreadability of free ITZ gel and ITZ-TFS gel was 4.5 ± 0.46 and 4.6 ± 0.29 cm, respectively. These results indicate the ability of prepared gels to be spread easily on the skin with low shear [ 51 ]. The pH value for the prepared gels were 6.62 ± 0.52 and 6.61 ± 0.74 for free ITZ gel and ITZ-TFS respectively.…”

“…The results show that including the vesicles in the gel improves the ability to control release and improve skin permeation. The Carbopol-based gel was used as a vehicle for transferosomes loaded with 5-FU [141], paclitaxel [142], methotrexate [143], tofacitinib [144], tamoxifen citrate [145], and others. An interesting study was presented by Shamim et al, which includes carvedilol in transferosomes for topical administration to prevent UV-induced skin cancer [146].…”

“…The test is performed on healthy volunteers with an established protocol of application. At the beginning and after one week of application the skin hydration is determined based on capacity measurements with specific probes of Soft Plus apparatus (Callegari Srl, Parma, Italy) [196,197] In vitro release This is an acellular assay in phosphate buffer with different pH [114] through a dialysis membrane [157] or with a membrane attached to a Franz-diffusion cell [137,143] which measures the amount of drug released over time.…”

Ex-vivo Skin Penetration StudyA Franz diffusion cell, with a 15.0 mL receptor compartment capacity and a 2.54 cm 2 diffusion area was used for the ABSTRACT Objective: The current study's objective is to assess the transfersomal gel, administration of tamoxifen citrate transdermally for Design of Experiments (DoE) Approach. Materials and Methods: Supramolecular clusters with extreme flexibility, known as transferosomes can penetrate untouched animal skin.
