-The objective in this research was to evaluate the isoflavone content and the physiological quality of seed from conventional and transgenic soybean cultivars before and after 180 days of storage. Twenty one soybean cultivars: CD 202, CD 206, CD 208, CD 213RR, CD 214RR, CD 215, CD 216, CD 217, CD 218, CD 221, BRS 184, BRS 185, BRS 214, BRS 244RR, BRS 245RR, BRS 246RR, BRS 255, BRS 257, BRS 258, BRS 261 and BRS 262, grown in the 2005/2006 crop season, were assayed. The seeds were packed in Kraft paper bags and stored at room temperature under laboratory conditions. Seeds were evaluated with respect to their germination and vigor (first germination count, accelerated aging and tetrazolium test) and their total isoflavone contents and respective aglycon forms (daidzein, genistein and glycitein),glycosides (daidzine, genistine and glycitine) and malonyl conjugates. A completely randomized block design with six replications with the treatments set out within a subplot scheme (21 cultivars x 2 storage periods) was used. The F-test was used to compare means between storage periods and the Scott-Knott test to compare cultivars for each storage period, both with a 95% probability. It was concluded that isoflavone contents differ between cultivars and show a distinct behavior throughout storage. isoflavonas totais e suas respetivas formas agliconas (daidzeína, genisteína e gliciteína), glicosídeos (daidzina, genistina e glicitina) e seus conjugados malonil. O delineamento experimental utilizado foi o inteiramente casualizado com 6 repetições e tratamentos dispostos no esquema de parcelas subdivididas (21 cultivares x 2 tempos de armazenamento). Para a comparação de médias entre tempos de armazenamento, utilizou-se o teste F e para comparações de cultivares para cada tempo de armazenamento utilizou-se o teste de agrupamento de Scott-Knott, ambos com 95% de probabilidade. Pelos resultados foi possível concluir que os teores de isoflavonas diferenciam-se entre as cultivares e possuem comportamento distinto ao longo do armazenamento.Termos para indexação: agliconas, Glycine max, daidzina, genistina, vigor.

RESUMO -Sementes de milho tornam-se tolerantes à dessecação à medida que secam naturalmente no campo e, após a maturidade fisiológica, pré-secagem a 35ºC pode induzir tolerância à temperatura de secagem de 50ºC. Nesse trabalho, sementes da cultivar BRS-3060, colhidas com teor de água de 42,2%, submetidas a períodos crescentes de pré-condicionamento apresentaram tolerância crescente à temperatura de 50ºC, até atingirem o teor de água de 25,9%, quando exibiram o máximo desempenho fisiológico, avaliado por meio de testes de germinação, vigor e atividade de enzimas α-amilase. O presente trabalho teve o objetivo de investigar o padrão eletroforético de enzimas removedoras de radicais livres e de proteínas lea, em sementes tolerantes e intolerantes a alta temperatura de secagem. As atividades das enzimas removedoras superóxido dismutase, peroxidase e catalase, foram detectadas em hipocótilos de plântulas após cinco dias de germinação e a atividade de proteínas lea detectada em eixos embrionários. Os resultados permitiram concluir que a tolerância de sementes de milho à temperatura de 50ºC está associada à atividade da enzima catalase e pouco relacionada à atividade das enzimas superóxido dismutase e peroxidase. Proteínas lea estavam ausentes em sementes de milho intolerantes e sua presença foi associada ao desempenho fisiológico das sementes tolerantes.Termos para indexação: Zea mays, tolerância à dessecação, peroxidase, superoxidase, catalase. FREE RADICAL-SCAVENGING ENZYMES AND LEA PROTEINS ASSOCIATED TO MAIZESEED TOLERANCE TO HIGH DRYING TEMPERATURE.ABSTRACT -Maize seeds assume desiccation tolerance as they dry naturally during maturation and pre drying at 35ºC after their physiological maturity can induce tolerance to artificial drying at 50ºC. Seeds of 'BRS-3060' harvested with 42,2% water content and submitted to increased periods of pre-conditioning, showed enhanced tolerance to 50ºC until 25,9% water content when they reached maximum physiological performance. Seed quality was evaluated by germination and vigor tests and α-amilase enzyme activity. Seeds tolerant and not-tolerant to drying at 50ºC were investigated by means of electrophoretic standards of free radical-scavenging enzymes and lea proteins. The scavenger activity enzyme superoxide dismutase, peroxidase and catalase was detected in 5-day old seedling hypocotyl and lea protein was detected in embryonic axes. The results showed that maize seeds tolerant to 50ºC are associated to catalase enzyme activity but are less related to superoxide dismutase and peroxidase enzymes activities. Lea proteins were not present in maize seeds non tolerant to desiccation and their presence was associated to the physiological performance of tolerant seeds.

ABSTRACT.Seeds collected at different maturation stages vary in physiological quality and patterns of protective antioxidant systems against deterioration. In this study we investigated the expression of genes that codify catalase (CAT), dismutase superoxide (SOD), and polyphenol oxidase (PPO) during the pre-and post-physiological maturation phases in whole seeds and in endosperms and embryos extracted from the seeds. Coffea arabica L. berries were collected at the green, yellowish-green, cherry, over-ripe, and dry stages, and the seeds were examined physiologically. The transcription levels of the genes were quantified by quantitative real-time polymerase chain reaction using coffee-specific primers. The highest level of SOD expression was observed in the endosperm at the cherry and over-ripe stages; in addition, these seeds presented the greatest physiological quality (assessed via germination test). The highest CAT3 transcript expression was observed at the green stage in whole seeds, and at the green and over-ripe stages in the embryos and endosperms. High expression of the PPO transcript was observed at the green and yellowish-green stages in whole seeds. In embryos and endosperms, peak expression of the PPO transcript was observed at the green stage; subsequently, peaks at the cherry and over-ripe stages were observed. We concluded that the expression patterns of the SOD and CAT3 transcripts were similar at the more advanced maturation stages, which corresponded to enhanced physiological seed quality. High expression of the PPO transcript at the over-ripe stage, also observed in the embryos and endosperms at the cherry stage, coincided with the highest physiological seed quality.
