Depolymerisation products of hyaluronic acid after exposure to oxygen-derived free radicals.

Abstract: SUMMARY Preparative chromatographic fractions of human umbilical cord hyaluronic acid (HA) of a molecular weight of 106 were subjected to graded oxygen-derived free radical (oxy radical) fluxes produced by: (a) the autoxidation of ferrous ions; (b) the action of xanthine oxidase (XO) on hypoxanthine (HX); and (c) by peripheral blood polymorphonuclear leucocytes that had been stimulated by phorbol myristate acetate (PMA). Analysis by gel chromatography of the products obtained with each of the oxy radical gener… Show more

“…Infants of HIV seropositive mothers are almost always seropositive at birth but might during the first 18 months of life turn seronegative if they are not infected (44) (49), and that this depolymerization is induced by oxygen free radicals (58,59), again fits in the previous hypothesis. This same approach, i.e., the evaluation of tissue LPO products levels in tissues, has been also applied to experimental models with similar results.…”

“…HA oligomers, and thus HA-degradation products, can have pro-or anti-inflammatory effects depending on their molecular weight [72][73][74]. Actually, different reactive oxygen species involved in the degradation process lead to different chemically modified HA oligomers, which are the ones involved in inflammation [75,76]. Low molecular weight fragments of HA tend to be pro-inflammatory, whereas high-molecular weight fragments have anti-inflammatory effects [77].…”

“…However, under inflammatory conditions HA has been shown to be more polydisperse, with a preponderance of lower molecular weight forms (4). The accumulation of lower molecular weight forms of HA has been postulated to occur by a variety of mechanisms including depolymerization by reactive oxygen species, enzymatic cleavage, and de novo synthesis of lower molecular weight species (3,(5)(6)(7)(8). Several studies have suggested that high and lower molecular weight HA may exhibit different biological effects on cells and in tissues (9,10).…”
