Deleterious effect of TRIS buffer on growth rates and pigment content of Gracilaria birdiae Plastino & E.C. Oliveira (Gracilariales, Rhodophyta)

Show more

“…They were transported to the laboratory and unialgal cultures were established from fragments, as described by Oliveira et al (1995). Cultures were maintained in Provasoli's enriched seawater (20 mL L −1 ; 31 psu salinity), which was prepared without Tris phosphate (Ursi et al 2008), in a temperaturecontrolled room at 9±1°C and 55 μmol photons m −2 s −1 PAR provided by Philips TLT 20 W/54 daylight fluorescent tubes, on a 12 h light/12 h dark cycle. The medium was renewed weekly.…”

“…Compared to wildtype, the light-green strain showed lower growth rates (Plastino et al 2004). The best culture conditions in the laboratory for growth and reproduction were provided (Ursi and Plastino 2001;Ursi et al 2008), making it possible to complete the life history of wild-type, greenish-brown, and green strains of G. birdiae in a laboratory culture . The photosynthetic, respiratory, and carotenoid concentrations were also investigated for these strains (Ursi et al 2003).…”

“…This species has recently been the focus of several laboratory studies (Plastino, 2004) and its life history has been completed in a laboratory culture (Costa & Plastino, 2001). Strain selection and the physiological demands of the strains were performed with the aim of improving cultivation techniques in the sea (Costa & Plastino, 2011;Ursi & Plastino, 2001;Ursi et al, 2003;Plastino et al, 2004;Ursi et al, 2008). Studies related to the cultivation of this species have been conducted in different environments such as shrimp ponds (Marinho-Soriano et al, 2009;Oliveira, 2007), estuaries (Marinho-Soriano et al, 2006) and the sea (Bezerra & Marinho-Soriano, 2010), showing promising growth rates.…”
