Delaying effect of cyclophosphamide on bone fracture healing chemical analyses of rat femora

Abstract: Hydroxyproline, calcium and inorganic phosphate contents of osteotomised rat femora were compared with those from rats treated with cyclophosphamide. The treatment consisted of 10 mg/kg/body weight injected intraperitoneally daily for 14 days after (group II) and before (group III) operation. The obtained results indicated a reduction in the hydroxyproline content of the callus and a diminution of the calcium and phosphate deposition upon it, thus inhibiting collagen formation and delaying mineralisation, reta… Show more

“…The goal of this study was to evaluate the effect of cisplatin on the formation of extracortical bone and periprosthetic soft tissue for a canine segmental re-placement model. The effects of different chemotherapeutic agents on bone formation have been studied in v i m and in small animals (8,15,29). Information about the effects of cisplatin on fracture healing (29), on bone turnover (2), and on allograft incorporation (34) has been reported, but there are no data concerning the formation of periprosthetic extracortical capsules, to our knowledge.…”

“…The time of drug administration seems to be a critical factor. Sommer-Tsilenis et al (29) administered cyclophosphamide daily for 14 days, beginning 16 days before or 2 days after midfemoral osteotomy. Although collagen deposition and mineralization of the fracture callus were significantly reduced for the animals treated postoperatively, an acceleration in bone healing was found for the animals treated preoperatively, such that these animals exhibited no significant differences from untreated controls.…”

“…Early descriptions of toxic skeletal side effects included osteoporosis, bone pain, and pathological fractures [ 16,211. Subsequent investigations have demonstrated a significant effect of both ADR and CDP on osteochondrogenesis in dog, rabbit, and rat models [10, 27,35]. These drugs appear to act specifically upon osteoblasts, with little effect on osteoclastic activity.…”
