Purpose Food-derived bioactive peptides may influence important physiological functions. An important example is beta-casomorphins, which are opioid peptides derived from A1 beta-casein in bovine milk and have been associated to be risk factors for non-communicable diseases in humans. A1 and A2 beta-casein are different with respect to the release of bioactive peptides, in particular BCM-7. However, evidence from human studies is limited and could be complemented with evidence derived from animal studies. We conducted a scoping review to identify animal studies investigating the effects of A1 beta-casein or BCM-7 compared to A2 beta-casein or any other intervention on health-related outcomes. Methods We systematically searched for relevant studies in two electronic databases (Medline, Embase; last search performed March 2020). Two reviewers independently undertook study selection and data extraction of included references. Results were summarized tabularly and narratively. Results We included 42 studies investigating various animal models, including rats, mice, rabbits, and dogs. Six studies investigated health-related outcomes of A1- vs. A2 milk, while most studies (n = 36) reported on physiological properties (e.g., analgesic effect) of BCM-7 as an opioid peptide. Included studies were extremely heterogeneous in terms of the study population, type of intervention and dose, and type of outcome measures. Conclusions Only a few studies comparing the effects of A1- and A2 milk were identified. More studies addressing this research question in animal models are needed to provide essential information to inform research gaps. Results from future studies could eventually complement research for humans, particularly when the body of evidence remains uncertain as is the case in the A1- and A2 milk debate.

BIOLOGICZNIE AKTYWNE PEPTYDY UWALNIANE Z BIAŁEK ŻYWNOŚCI S t r e s z c z e n i eBiałka żywności charakteryzują się wieloma właściwościami odżywczymi i biologicznymi. Biologicznie aktywne peptydy to fragmenty sekwencji aminokwasowych białek żywności, które stają się aktywne po uwolnieniu. Zwykle są one uwalniane podczas procesów trawienia, fermentacji (dzięki aktywności proteolitycznej mikroorganizmów) lub procesów enzymatycznych in vitro i wówczas mogą wpływać na zdrowie człowieka. Z białek żywności wyizolowano szereg peptydów bioaktywnych, w tym: inhibitory enzymu konwertującego angiotensynę, antyoksydacyjne, antymikrobiologiczne, antyamnezyjne, opioidowe, sensoryczne czy wiążące mikroelementy. Badane są także peptydy niekorzystnie oddziałujące na zdrowie człowieka, np. toksyczne dla osób chorych na celiakię. Obecnie kontynuowane są badania w celu wskazania nowych źródeł bioaktywnych peptydów, a także sposobów ich otrzymywania, biodostępności, aktywności biologicznej i mechanizmów działania. W artykule przedyskutowano sposoby otrzymywania bioaktywnych peptydów z białek żywności, wybrane rodzaje ich aktywności biologicznej oraz ich biodostępność.Słowa kluczowe: białka żywności, bioaktywne peptydy, struktura bioaktywnych peptydów, biodostępność WprowadzenieŻywność konwencjonalną rozpatruje się obecnie nie tylko jako źródło składników odżywczych niezbędnych do utrzymania homeostazy organizmu, ale również jako źródło składników bioaktywnych. Dowiedziono bowiem, że białka żywności są źró-dłem bioaktywnych peptydów, które mogą regulować procesy fizjologiczne ustroju, w tym wszystkich układów organizmu, np. hormonalnego, immunologicznego, krąże-nia, nerwowego czy pokarmowego, co wykorzystuje się obecnie m.in. w produkcji Prof. dr hab. β-Casomorphins are a group of opioid peptides released during gastrointestinal digestion or food processing from the β-casein of milk protein. Consequently, milk can be divided into A1 and A2 "like" groups depending upon the presence or absence of proline or histidine at the 67th position of β-casein. A1 "like" milk is postulated to be a source of BCM-7 as histidine allows the cleavage at this position, while A2 "like" milk has proline that resists the hydrolysis. On one hand, BCM-7 has been implicated as a risk factor for cardiovascular diseases, type I diabetes, and neurological disorders. On the other hand, various physiological effects of these peptides have also been documented, i.e., secretion of mucus, increased activity of superoxide dismutase and catalase, increased levels of prolactin, and analgesic role. In addition, many evidences correlate these peptides with various immunological functions, such as development of innate immunity, lymphocyte proliferation and cellular immunity, role in autoimmune diseases, histamine release, and allergy. In conclusion, the role of β-casomorphins in physiological functions remains controversial and more research with improved diagnostic techniques is needed to unravel the mechanism and study physiological functions of β-casomorphins. Thus, health-related aspects of β-casomorphins (positive, negative, and immunological impacts) have been comprehensively reviewed in this article.
