Decursin Alleviates Mechanical Allodynia in a Paclitaxel-Induced Neuropathic Pain Mouse Model

Abstract: Chemotherapy-induced neuropathic pain (CINP) is a severe adverse effect of platinum- and taxane-derived anticancer drugs. The pathophysiology of CINP includes damage to neuronal networks and dysregulation of signal transduction due to abnormal Ca2+ levels. Therefore, methods that aid the recovery of neuronal networks could represent a potential treatment for CINP. We developed a mouse model of paclitaxel-induced peripheral neuropathy, representing CINP, to examine whether intrathecal injection of decursin coul… Show more

“…The findings were determined to be in line with those of previous research. Numerous studies have concentrated their attention on the neuronal level of the sciatic nerve and spinal cord in paclitaxel-induced neuropathy and have found morphological abnormalities at both levels [ 49 ]. In addition, we investigated the morphology of two different tissues and came to the conclusion that PTX played a role in the progression of the disease by mediating neuronal degeneration, disrupting the development of myelin sheaths, and causing vacuolation.…”

“…10. One of the important aspects of paclitaxelinduced neuropathy is the disturbance in latency of falling [40], body weight [41], and mechanical allodynia [42]. The behavioral analysis of our compounds reveals that berbamine, bergepten and carveol are corrected the behavioral de cit in paclitaxel-induced peripheral neuropathic pain as is shown in Fig.…”

“…Accumulating evidence suggests that phytochemicals can affect the central nervous system by crossing the BBB. Thus, these phytochemicals may have played a role in the analgesic effect of JI017 as decursin has been reported to attenuate paclitaxel-induced pain and to act as an TRPV1 antagonist [65], while aconitine has also been shown to have an analgesic effect on various animal models of pain [66]. The analgesic effect of 6-gingerol was also reported in several studies [67][68][69].…”
