Decreased Expression of Stem Cell Markers by Simvastatin in 7,12-dimethylbenz(a)anthracene (DMBA)–induced Breast Cancer

Abstract: Simvastatin, a competitive inhibitor of HMG-CoA reductase widely used in the treatment and prevention of hyperlipidemia-related diseases, has recently been associated to in vitro anticancer stem cell (CSC) actions. However, these effects have not been confirmed in vivo. To assess in vivo anti-CSC effects of simvastatin, female Sprague-Dawley rats with 7,12-dimethyl-benz(a)anthracene (DMBA)-induced mammary cancer and control animals were treated for 14 days with either simvastatin (20 or 40 mg/kg/day) or soybea… Show more

“…ALDH1A1 activity is also associated with ERK→C/EBPβ [64] and NOTCH [65] signaling. With reference to our study, CD24, CD44, EpCam, and ALDH1A1 have been previously proven to be good CSC markers in chemocarcinogen-induced rat carcinogenesis [66,67]. This study aimed to characterize T. vulgaris effects on the expression of CSC-like markers in the chemoprevention model of breast carcinogenesis and a significant decrease in CD44 and ALH1A1 expression in rat mammary tumors was found.…”

“…The phenotype of CD44 + /CD24 -/low and overexpression of ALDH1, EpCAM, and nestin suggest a worsened prognosis of BC in women [74]. CD24, CD44, EpCAM, and ALDH1 have been shown to be good markers of CSCs also in chemically induced mammary carcinogenesis in rats [75,76]. Based on comparative preclinical oncology studies, plant functional foods (containing mixtures of phytochemicals) are supposed to exhibit better anticancer activities (including the anti-CSC properties) when compared to isolated phytochemicals [1].…”

“…Currently, the evaluation of CD44, CD24 and ALDH1 expressions is considered as the most precise methodology to identify CSCs in the breast cancer women . Recently, both CD24 and CD44 have been proven as good CSCs markers in chemocarcinogen‐induced rat mammary tumorigenesis . Lower expression of CD24, CD44, ALDH1 and EpCAM is linked with better prognosis of breast carcinoma in patients .…”
