Decreased expression of L‐selectin (CD62L) on lymphocytes in enzootic bovine leukaemia

Abstract: Expression of L-selectin was determined by single- and two-colour immunofluorescence on granulocytes, peripheral blood mononuclear cells (PBMC) and blasts of bovine origin by means of a monoclonal antibody IVA94 which recognizes bovine L-selectin (CD62L). Cells were separated from peripheral blood of healthy cattle and colleagues infected with bovine leukaemia virus (BLV). BLV-infected animals comprised lymphocytotic and non-lymphocytotic cows. L-selectin was expressed on 90-98% of granulocytes in all tested a… Show more

“…In the current study, we show a reduction in CD62L expression that is most pronounced 2 d following infection, followed by a rapid recovery of CD62L expression, similar to what has been reported during bovine leukemia virus (26) or Mycobacterium bovis (27) infection. In contrast to our finding in this viral infection, Walcheck and Jutila (28) found a much slower downregulation of L-selectin following short-term activation of gd T cells in vitro with PMA.…”

“…Cows categorized as nLC or LPVL had the lowest average IgA and IgM concentrations in both milk and saliva. This result agrees with other investigators who report that BLV infection results in immune disruption even in cows with an aleukemic profile [ 33 , 34 ]. One potential mechanism for BLV-related interference with immunoglobulins is inhibition of the J-chain [ 35 , 36 ], which is essential for the assembly of IgA and IgM polymers as well as for the process of cross-membrane transport [ 37 , 38 ].…”

“…Cattle have decreased expression of CD62L when infected with Mycobacterium avium ssp. paratuberculosis (MAP; Waters et al, 2003), bovine respiratory syncytial virus (McInnes et al, 1999), bovine leukemia virus (Dusinsky et al, 2000), or foot and mouth disease virus (Toka et al, 2011). Therefore, CFC-fed calves could have a decreased ability to clear pathogens, resulting in shedding of CD62L from lymphocytes.…”
