BackgroundIn Kazakhstan, traditional artisanal cheeses have a long history and are widely consumed. The unique characteristics of local artisanal cheeses are almost completely preserved. However, their microbial communities have rarely been reported. The current study firstly generated the Single Molecule, Real-Time (SMRT) sequencing bacterial diversity profiles of 6 traditional artisanal cheese samples of Kazakhstan origin, followed by comparatively analyzed the microbiota composition between the current dataset and those from cheeses originated from Belgium, Russian Republic of Kalmykia (Kalmykia) and Italy.ResultsAcross the Kazakhstan cheese samples, a total of 238 bacterial species belonging to 14 phyla and 140 genera were identified. Lactococcus lactis (28.93%), Lactobacillus helveticus (26.43%), Streptococcus thermophilus (12.18%) and Lactobacillus delbrueckii (12.15%) were the dominant bacterial species for these samples. To further evaluate the cheese bacterial diversity of Kazakhstan cheeses in comparison with those from other geographic origins, 16S rRNA datasets of 36 artisanal cheeses from Belgium, Russian Republic of Kalmykia (Kalmykia) and Italy were retrieved from public databases. The cheese bacterial microbiota communities were largely different across sample origins. By principal coordinate analysis (PCoA) and multivariate analysis of variance (MANOVA), the structure of the Kazakhstan artisanal cheese samples was found to be different from those of the other geographic origins. Furthermore, the redundancy analysis (RDA) identified 16 bacterial OTUs as the key variables responsible for such microbiota structural difference.ConclusionOur results together suggest that the diversity of bacterial communities in different groups is stratified by geographic region. This study does not only provide novel information on the bacterial microbiota of traditional artisanal cheese of Kazakhstan at species level, but also interesting insights into the bacterial diversity of artisanal cheeses of various geographical origins.Electronic supplementary materialThe online version of this article (doi:10.1186/s12866-016-0911-4) contains supplementary material, which is available to authorized users.

Natural growth-promoting nutrients extracted from aquaculture sludge waste can be used to maximise microalgal growth. This study identified the influence of aquaculture sludge extract (SE) on four microalgae species. Conway or Bold's Basal Media (BBM) was supplemented with SE collected from a Sabak Bernam shrimp pond (SB) and Kota Puteri fish pond (KP), and tested using a novel microplate-incubation technique. Five different autoclave extraction treatment parameters were assessed for both collected SE, i.e., 1-h at 105 °C, 2-h at 105 °C, 1-h at 121 °C, 2-h at 121 °C, and 24-h at room temperature (natural extraction). Microalgae culture in the microplates containing control (media) and enriched (media + SE) samples were incubated for nine days, at 25 °C with the light intensity of 33.75 μmol photons m −2 s −1 at 12-h light/dark cycle. The total dissolved nitrogen (TDN) and total dissolved phosphorus (TDP) in KP SE were 44.0–82.0 mg L −1 and 0.96–8.60 mg L −1 . TDN (8.0%–515.0%) and TDP (105%–186 %) were relatively higher in KP SE compared to SB SE. The growth of microalgae species Nannochloropsis ocenica showed significant differences ( p < 0.05) between the five extraction treatments from SB and the control. However, Chlorella vulgaris , Neochloris conjuncta , and Nephroclamys subsolitaria showed no significant differences ( p > 0.05) in SB SE. N. ocenica , C. vulgaris , and N. conjuncta showed significant differences ( p < 0.05) between five extraction treatments from KP and the control while N. subsolitaria showed no significant difference ( p > 0.05). The specific growth rate (SGR) in the exponential phase of all microalgae species were relatively higher in SB SE compared to KP SE. While the organic matter content of KP SE was relatively higher, there were no significant differences in microalgae growth compared to SB SE. Nonetheless, modified SE did influence microalgae growth compared to the control. This study shows that modified SE could be used as enrichment media for microalgae cultivation.

The fate and transport of polychlorinated biphenyls (PCBs), a class of persistent organic compounds, in soils was markedly affected by their sorption/desorption on soil organic matters (SOM) due to high hydrophobicity of PCBs. Humic acid (HA), an important fraction of SOM, has no steady composition and microstructure from different origins, resulting in their diverse sorption capacity. Therefore, the effect of composition and microstructure of HA on sorption of 3,3',4,4'-tetrachlorobiphenyl (PCB77) was investigated in this study. The primary sorption mechanism of PCB77 on HAs was also demonstrated by using Fourier transform infrared (FTIR) and solid-state carbon-13 nuclear magnetic resonance (C-NMR) spectroscopy analysis. Elemental content analysis of three HAs showed that the content of unsaturated C (the total of aromatic, carboxylic, and carbonyl carbon) followed an order of YHA>SHA>AHA. Furthermore, YHA and SHA had remarkably rough and complicated particle surfaces but AHA had relatively smooth surface and the well-proportioned and interspersed particle sizes. The results of the sorption experiment showed that the sorption capacity of PCB77 on HAs followed a similar order of YHA>SHA>AHA, indicating the content of unsaturated C of HAs controlled the sorption of PCB77 on HAs. Sorption of PCB77 on either AHA or SHA did not change with increasing ionic strength of background solution, implying that there was no H-bond or electrostatic interaction between PCB77 and HAs. The result of FTIR and C-NMR spectra showed the primarily possible mechanism was π-π conjugative interaction and hydrophobic binding between PCB77 and HAs.
