Decipinin A and Decipienolides A and B:  New Bioactive Metabolites from the Coprophilous Fungus Podospora decipiens

Abstract: Decipinin A (1), a new compound with antifungal and antibacterial activity, has been isolated from liquid cultures of the coprophilous fungus Podospora decipiens (JS 270). Two new tetracyclic sesquiterpene lactones, decipienolides A (2) and B (3), were also obtained from this isolate as an inseparable mixture of epimers that showed antibacterial activity. The structures of 1-3 were elucidated by analysis of 1D and 2D NMR data, aided by chemical shift comparisons to related compounds.

“…Typically, C(1), C(2), C(3), and C(4) were À0.4, +1.1, À0.7, and +1.4 ppm shifted, which was in accordance with the deviactions between expansolides A (3) and B (4), [11] as well as decipienolides A and B. [12] Compounds 1 and 2 were named as expansolides C (1) and D (2), respectively.…”

“…The key HMBCs between CH 2 (14) and C(4) linked the oxygenated CH 2 (14) group to C(4) to form a tetrahydrofuran ring. The C=O group was determined to be C(1) by resorting to the HMBCs from H–C(2), CH 2 (3), and Me(13) to the resonance at δ (C) 181.3, while C(1) was linked to C(4) to form a γ ‐lactone ring, as indicated by the molecular formula, the chemical shift of C(1), as well as the IR absorption at 1745 cm −1 . Thus, the planar structure of 1 was established.…”

“…Phomadecalin C was also isolated from cultures of Phoma sp. (NRRL 25697), a fungus originally isolated from wood decay stromata (Che et al, 2002). Phomadecalin C was shown to be antagonistic to Bacillus subtilis (ATCC 6051) in standard disk assays (Che et al, 2002).…”
