“…Although there was about a twofold increase in the activity of this enzyme at the implantation sites between days 7 and 8, the main increase took place in the decidual nodules confirming previous findings (18,19,27) of a 10-fold increase in alkaline phosphatase activity on day 9 of pregnancy in the rat uterus, an increase localized exclusively to the implantation sites presumably containing the decidua. The biochemical findings also support the histochemical demonstra tions of increased alkaline phosphatase activity associated with decidualization (3,15,23).…”

“…It is unclear why these inhibitors did not prevent the ALP rise because the present studies show that there was a clear elevation of ALP mRNA correlating with the induction of ALP activity. 5-Flurodeoxyuridine and inhibitors of protein synthesis that inhibit DNA synthe¬ sis as a consequence of their actions also failed to reduce the decidual response in the studies of Manning et al (1969), despite the fact that decidualization involves substantial cell proliferation (Hall, 1969;Das & Martin, 1978). No measurements of the degree of inhibition of protein or RNA synthesis were performed by Manning et ai (1969).…”

“…The induction of enzyme activity was pre¬ ceded by an elevation in a 2-6 kb ALP mRNA which reached a peak 8-fold over the control level at Day 6. Manning et al (1969) were unable to inhibit the elevation of ALP-specific activity induced by traumatizing progesterone-primed rat uteri with inhibitors of transcription (actinomycin-D), replication (5-flurodeoxyuridine) or translation (cycloheximide, chloramphenicol). This led these workers to postulate that the ALP induction was due to post-translational activation of a pre¬ existing pro-enzyme.…”

“…An increase in alkaline phosphatase activity in the endometrial stroma is associated with decidualization (Lobel et al, 1965) and early pregnancy (Christie, 1966;Manning et al, 1969). This increase is maximal 24-72 h after the induction of decidualization and provides a good index of endometrial differentiation.…”
