De novo transcriptome sequencing in Frankliniella occidentalis to identify genes involved in plant virus transmission and insecticide resistance

Abstract: The western flower thrips (WFT), Frankliniella occidentalis, a world-wide invasive insect, causes agricultural damage by directly feeding and by indirectly vectoring Tospoviruses, such as Tomato spotted wilt virus (TSWV). We characterized the transcriptome of WFT and analyzed global gene expression of WFT response to TSWV infection using Illumina sequencing platform. We compiled 59,932 unigenes, and identified 36,339 unigenes by similarity analysis against public databases, most of which were annotated using g… Show more

“…Interestingly, among the differentially expressed genes, many genes related to cellular and humoral immune responses had higher transcript levels in the M population than in the TN1 population. Lysosomes and phagosomes have been shown to play a direct antiviral role [46]. In the M population, 38 genes (79.17%) involved in lysosome functions and 75 genes (85.23%) involved in phagosome functions had higher levels of transcripts compared to those in the TN1 population, strongly suggesting that these pathways were activated in the virulent M population.…”

“…The KEGG database contains data from a systematic analysis of inner-cell metabolic pathways and functions of gene products, in addition to variants specific to particular organisms 30 . Pathway-based analyses can help to further understand the biological function and expression level of genes 14 .…”

“…A draft genome, and transcriptome and proteome data for whole body and different developmental stages of F. occidentalis are available, and differentially expressed (DE) genes in response to TSWV infection including antiviral defense are being annotated [66]. Global gene expression in response to TSWV infection identified DE genes in cellular processes and innate immune response [173]. Primary salivary gland transcriptome data identified genes involved in detoxification and inhibition of plant defense responses, and genes that may play a role in the extra-oral digestion of plant tissues and sugars [91].…”
