Darnell Reconceptualized

Abstract: Darnell's recent article is neither a reconceptualization nor for that matter a conceptualization, and that there is in it in fact nothing new. Included are examples that should send a critical reader back to Darnell and back to the problem he raises-the adequacy of present theoretical formulations in our field.Darnell's recent article (1971a) in this journal presents certain special problems to the critic. It is carried in a scholarly journal, yet he asks us to consider it neither in the context of science no… Show more

“…The existence of rhodanese in plants has been highly controversial and only demonstrated recently in Arabidopsis after EST mining and subsequent characterization of cDNAs and recombinant proteins [40][41][42]. Although the exact function of thiosulfate sulfur transferase remains unclear, it has been postulated to play a role in several mechanisms such as the formation of iron-sulfur centers, sulfur assimilation and cyanide detoxification [76][77][78]. TST abundance in pea seed mitochondria is rather intriguing and will require in-depth investigation to elucidate its actual function in relation to germination.…”

“…Sulfurtransferase activity was measured by the method of Westley (17). Reaction mixtures contained 100 mM Tris-acetate, 50 mM ammonium thiosulfate, and 50 mM KCN (pH 8.6) in a volume of 500 l. After 5-30 min, formaldehyde (250 l, 15%) was added to quench the reaction, and color was developed by the addition of 750 l of ferric nitrate reagent [100 g of Fe(NO 3 ) 3 ⅐9H 2 O and 200 ml of 65% HNO 3 per 1,500 ml].…”

“…The optimum pH for rhodanese activity is 8.6 (8). In these studies the activity was measured at pH 8.6 and at 7.0.…”
