The cytotoxicity of nine soft and hard lining materials (Mollosil Plus, Ufi Gel SC, Visco-gel, Molloplast-B, GC Tissue Conditioner, Vertex Rapid Simplified, GC Reline Hard, Vertex Self-Curing, Ufi Gel hard C) was evaluated using human gingival fibroblasts (HGFs). Twelve disk samples per lining material were prepared and incubated for 24, 48, 72, and 96 h. Cytotoxicity of each lining material's extract on cultured HGFs was measured using XTT assay. Data were analyzed using one-way ANOVA, post hoc Dunnett's T3 and Bonferroni tests at a significance level of p<0.05. At all incubation periods, all the hard lining materials (Vertex-SC, GC Reline Hard, Vertex-RS, and Ufi Gel hard C) showed cell viability higher than 90%. Among the soft lining materials, although there were no significant differences in cell viability among the different incubation periods for each lining material (p>0.05), autopolymerized acrylic-based GC Tissue Conditioner showed significantly lower cell viability than the other soft lining materials at each incubation period. Among the hard lining materials, there were no significant differences both among the materials and across all incubation periods for each lining material (p>0.05). In conclusion, all soft and hard liners exhibited good biocompatibility regardless of incubation time, except for GC Tissue Conditioner.

Influence of plaster drying on the amount of residual monomer in heat-cured acrylic resins

Aim: To evaluate the influence of plaster condition, dry or not, on the amount of residual monomer in heat-cured acrylic resin. Methods: Thirty acrylic resin specimens (65x10x3 mm) were fabricated and randomly assigned to 5 groups (n=6). The evaluated resins were heat-cured acrylic resins by conventional or microwave polymerization techniques and the plaster was previously dried in microwave oven in two groups. Each specimen was individually immersed in a test tube containing methanol (7 days) for surface analysis. In the groups for which internal monomer was evaluated, the specimens were fragmented and the small fragments were weighed prior to immersion in methanol. The analysis was made by high performance liquid chromatography (HPLC). Data were analyzed by ANOVA and Tukey test (p<5%) Results: showed statistical differences among the groups. Conclusions: The previous plaster drying influenced the residual monomer amount showing a decrease of these levels.

ÖZAmaç: Çalışmada, iki farklı yumuşak astar materyalinin suda 3 farklı yaşlandırma süresi sonrasındaki sitotoksik özelliklerinin karşılaştırmalı olarak değerlendirilmesi amaçlanmıştır. Gereç ve Yöntemler:162 adet 5 mm çapında ve 2 mm kalınlığında 2 farklı yumuşak astar materyalinden (Grup A: Hydrocast; n=81, Grup B: Trusoft; n=81) hazırlanan disk örnekler 3 alt gruba ayrıldı. Grup I: 1 gün suda yaşlandırma, Grup II: 2 gün suda yaşlandırma, Grup III: 7 gün suda yaşlandırma. Belirtilen yaşlandırma sürelerini takiben fare fibroblast hücreleri (L929) kullanılarak; 24 saat, 48 saat ve 72 saat hücre inkübasyonu sonrasında her grup için ortalama hücre canlılık yüzdeleri 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide testi ile belirlendi. Gruplar arasındaki istatistiksel farklılıkların tespiti için ANOVA (One-Way) testi kullanıldı. (p<0,05). Bulgular:En düşük hücre canlılık oranları 24 saat hücre inkübasyonu sonrasında gözlendi. 24 saat hücre inkübasyonunu takiben 1 gün ve 7 gün suda bekletilen Hydro-cast örnekler ve 7 gün suda bekletilen Trusoft örnekler çok az sitotoksik özellik gösterdi. Diğer gruplarda sitotoksik özellik belirlenmedi.Sonuç: Farklı suda yaşlandırma süreleri sonrasında kullanılan yumuşak astar materyallerinin sitotoksik özellikleri değişebilmektedir.Anahtar Sözcükler: Suda yaşlandırma, Sitotoksisite, Yumuşak astar, 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide testi ABSTRACT Objective: The purpose of this study was to examine the cytotoxicity of 2 soft relining materials following 3 different aging periods.Material and Methods: 162 disc specimens of 5 mm radius and 2 mm thickness and made of 2 different soft lining materials (Group A: Hydrocast; n=81, Group B: Trusoft; n=81) were divided into 3 subgroups (Group I: 1 day water aging, Group II: 2 days water aging , Group III: 7 days water aging). After the water aging periods, the cytotoxicity of the lining materials was determined by using mouse fibroblast (L929) cells incubated with 3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide after 24 hours, 48 hours and 72 hours. Statistical differences were analysed with ANOVA (One-way) (p<0.05).Results: Lowest cell vitality values were observed after the 24-hour cell incubation period. Both Hydrocast and Trusoft samples stored in water for 7 days and Trusoft samples stored in water for 24 hours showed low cytotoxixity after the 24-hour cell incubation period. Conclusion:The aging period is a factor that influences the cytotoxic characteristics of soft relining materials.
