Cytotoxic T-Lymphocyte Activity Specific for Hemagglutinin (H) Protein of Canine Distemper Virus in Dogs.

Abstract: ABSTRACT. Cytotoxic T-lymphocyte (CTL) responses to hemagglutinin (H) protein of canine distemper virus (CDV) were evaluated in dogs using the replication-deficient adenovirus protein expression system. Skin fibroblasts were isolated from two dogs and were infected with recombinant adenovirus bearing the CDV-H gene (Ade-CDVH). CTL assay was performed using fibroblasts expressing CDV-H protein as target cells and peripheral blood lymphocytes (PBL) collected from the same dogs one week after immunization of CDV … Show more

“…The L, N and P proteins, together with a viral RNA, constitute the ribonucleic protein (RNP). The H and F glycoproteins facilitate viral entry process by binding and fusing to the host cells, respectively 2,3 . The H protein is largely known to have the antigenic variation and the antibodies to H protein are critical for the protective immunity against infection 4 .…”

“…[30][31][32] To determine whether mRNA-loaded canine CD40-B cells can induce functional T-cell responses in vitro against a viral antigen, cCD40-B cells were generated from three privately owned healthy dogs that had previously received the attenuated live CDV vaccine (Fort Dodge Animal Health, Kansas, IA, USA). cCD40-B cells harvested on day 14 of culture were electroporated with CDV-HA mRNA and co-cultured with autologous PBMC in the presence of rhIL-2 and rhIL-7.…”

“…The construction and characterization of the first replication-competent rCAd2 started recently. The genes that code for CDV HA 28,39 or fusion (F) 28 proteins were inserted in two CAd2s and used as a candidate vaccine in puppies. It was reported that intranasal vaccination with a mixture containing 10 5.8 TCID 50 of which rCAd2 provided an excellent level of protection in seronegative puppies, inducing almost complete protection.…”
