Cytotoxic, apoptotic, and genetic evaluations of Nigella sativa essential oil nanoemulsion against human hepatocellular carcinoma cell lines

Abstract: Background Phytochemicals and plant extracts are showing promising anticancer potentials. In the current study, the volatile faction (essential oil) of Nigella sativa seeds was evaluated against some hepatocellular carcinoma (HCC). The essential oil was extracted and characterized by chromatographic techniques to reveal its chemical composition, especially thymoquinone. Then, the oil was fabricated in two nanoemulsion formulations (F1 and F2), which differ in their composition of surfactants. T… Show more

“…The nucleic acid sequences of the forward (F) and reverse (R) primers of Bax and Bcl-2 genes compared with GAPDH, as a housekeeping gene, were shown as designed in recent Abd-Rabou's published paper. [33] Real-time PCR mixture consisted of 10 μL of SYBR PCR Master Mix, 1 μL of each primer (400 nM), 1 μL cDNA, and 7 μL Rnase-free water in a total volume of 20 μL. Amplification conditions and cycle counts were a temperature of 95 C for 15 min for the initial activation, followed by 40 cycles of denaturation at 95 C for 30 s, annealing at 55 C for 30 s, and elongation at 72 C for 30 s. Melting curves were performed after real-time PCR to demonstrate the specific amplification of each gene versus GAPDH as an internal control to normalize the level of target gene expression.…”

“…Abd-Rabou noted that nanoemulsions of Nigella sativa-EO inhibited the proliferation of hepatocellular carcinoma cells more effectively than pure EO after a 24-hour incubation. Speci cally, these nanoemulsions demonstrated inhibition rates of ~ 29% and ~ 15% against HepG2 and Huh-7 cells, respectively 57 . Such antiproliferative effects are likely linked to the presence of terpinen-4-ol 58 and cinnamaldehyde 59 , key chemical components found in TEO and CEO.…”

“…It has crucial signaling functions in physiological and pathological processes such as brain signaling, immunological responses, and blood pressure regulation [79]. Excessive nitric oxide production has been linked to various harmful illnesses, including trauma, dermatitis, and cancer [80]. The nitric oxide scavenging activity of the synthesized AgNPs is given in Figure 6.…”
