Cytosolic and mitochondrial ROS: which one is associated with poor chromatin remodeling?

Abstract: The aim of this study was to determine if there are any associations between impaired chromatin packaging and the origin of reactive oxygen species (ROS) production. Cytosolic ROS, mitochondrial ROS, DNA protamination, and apoptosis were evaluated with dichlorofluoresceindiacetate (DCFH-DA), dihydrorhodamine 123 (DHR123), chromomycin A3 (CMA3), and YO-Pro-1 (Y1)/propidium iodide (PI), respectively, by flow cytometry (FCM) in 40 infertile individuals. Percentages DCF+ and R123+ sperm were positively associated … Show more

“…the efficiency of chromatin remodeling in the male line is in keeping with previous publications demonstrating that strong relationships exist between the efficiency of chromatin protamination, as measured by chromomycin 3 (CMA3) fluorescence, and both ROS generation and oxidative DNA damage-the authors of this paper cannot claim to be the first to make this link [8,9]. 4.…”

“…Poor protamination with elevated levels of peripheral histone retention would leave the chromatin far less dense and exposed to ROS attack [Hammoud et al 2011]. In fact, a positive correlation was found between poor protamination and higher levels of ROS production and subsequent DNA fragmentation in individuals with abnormal semen parameters [Kiani-Esfahani et al 2013]. Besides, the retention of histone is not random and is known to have epigenetic significance making it one of the important parameters to be studied in sperm of RPL [Brunner et al 2014;Castillo et al 2014].…”

“…Studies have demonstrated that inappropriate culture mediums increase ROS resulting in OS events, DNA damage and low quality of oocytes. These are the main factors that lead to failure in IVM and decrease productivity [9,10]. OS appears to be responsible for many injuries in the embryo and oocyte [9,11,12].…”
