Large tumor suppressor kinase 2 (LATS2) is a core component in the Hippo signaling pathway, and it functions as a tumor suppressor associated with tumor cell proliferation and apoptosis. The purpose of this study is to explore LATS2 expression and its clinicopathological significance in non-small cell lung cancer (NSCLC). We examined LATS2 protein expression by immunohistochemistry in 184 resected NSCLC specimens using tissue microarrays. Low LATS2 expression was significantly related to disease recurrence (p = 0.047). In survival analysis, the low LATS2 expression group showed a statistically poorer overall survival (OS) (p = 0.004) and disease-free survival (DFS) (p = 0.014) than the high expression group. In multivariate analysis, downregulated LATS2 expression in NSCLC could be an independent prognostic factor of poor OS and DFS. Furthermore, we evaluated the prognostic significance of LATS2 expression in two major NSCLC subtypes, squamous cell carcinoma and adenocarcinoma. The low LATS2 expression group showed worse prognosis than the high LATS2 expression group (OS [p = 0.144] and DFS [p = 0.022] in squamous cell carcinoma and OS [p = 0.045] and DFS [p = 0.271] in adenocarcinoma). We demonstrated that downregulated LATS2 expression may predict aggressive biologic behavior and a worse prognosis in NSCLC and we also suggested the possibility of LATS2 as a therapeutic target in both squamous cell carcinoma and adenocarcinoma.

LINC00355 has been reported aberrantly over-expressed and associated with poor prognosis in various types of cancer. However, reports regarding the effect of LINC00355 on lung squamous cell carcinoma (SCC) are rare. This study aimed to explore the function of LINC00355 in the development and progression of lung SCC and reveal the underlying mechanism. The expression and subcellular location of LINC00355 were determined by qRT-PCR and RNA-FISH, respectively. The lung SCC cell growth was analyzed by CCK-8 assay, transwell invasion, wound healing, colony formation, and flow cytometry assays. Reactive oxygen species level was evaluated by DCFH-DA probes. Bioinformatics online websites, luciferase reporter assay, RNA binding protein immunoprecipitation (RIP), and RNA pull-down assays were utilized to investigate the interaction among LINC00355, miR-466, and Ly-1 antibody reactive clone (LYAR). The results showed that LINC00355 was upregulated in lung SCC and was positively associated with poor overall survival in lung SCC patients. LINC00355 was mainly located in the cytoplasm of SCC cells. Additionally, LINC0035 functioned as a competing endogenous RNA (ceRNA) to target miR-466, and LYAR was identified as a direct target of miR-466. LINC00355 expression negatively correlated with miR-466 level, and positively correlated with LYAR level. Mechanistically, knockdown of LINC00355 inhibited cell proliferation, migration and invasion, promoted cell apoptosis in vitro , and suppressed tumor growth in vivo through targeting miR-466, and thus down-regulated LYAR expression. These findings provide a new sight for understanding the molecular mechanism of lung SCC and indicate that LINC00355 may serve as a potential biomarker for the diagnosis and treatment of lung SCC.

Objective To explore the association of maspin expression with clinical pathological parameters and prognosis in non-small cell lung cancer (NSCLC) who received the surgical therapy. Methods The EMBASE, Web of Science and PubMed were searched to identify eligible studies to 3 June, 2019. The correlation of maspin expression with clinicopathological characteristics and survival of resected NSCLC patients was assessed by the combined relative risk (RR) and hazard ratio (HR) with corresponding 95% confidence interval (CI), respectively. All statistical analyses were performed via the Stata 12.0 software. Results A total of 12 articles involving 1771 patients were included. The results manifested that maspin was more prevalent in lung squamous cell carcinoma (SCC) (RR=0.36, 95% CI: 0.22-0.60, P <0.001) and significantly associated with P53 expression (RR=1.50, 95% CI: 1.00-2.24, P =0.048), while no significant correlation of maspin with other clinicopathological parameters such as the gender, age, tumor size, lymph node metastasis, tumor node metastasis (TNM) stage or differentiation status was observed. As for the prognosis representing as overall survival (OS), disease-free survival (DFS) and cancer-specific survival (CSS), only significant association between maspin and CSS (HR=1.58, 95% CI: 1.16-2.15, P =0.003) was revealed. However, the subgroup analysis for OS based on the histology demonstrated that maspin expression was an unfavourable and favourable prognostic indicator in lung adenocarcinoma (HR=3.36, 95% CI: 1.44-7.87, P =0.005) and SCC (HR=0.44, 95% CI: 0.27-0.71, P =0.001), respectively. Conclusion Maspin expression was correlated with histology type and P53 expression, but no certain association of maspin with other clinicopathological characteristics or prognosis was observed in resected NSCLC. More well-designed prospective researches with big samples are still needed to further assess the clinicopathological and prognostic significance of maspin in NSCLC patients undergoing surgical resection.
