Cytological staining of protozoa: a case study on the impregnation of hypotrichs (Ciliophora: spirotrichea) using laboratory-synthesized protargol

Abstract: Protargol (silver proteinate) impregnation is a common method used to identify and characterize ciliated protozoa. Unfortunately, chemical companies have stopped producing the 'strong' protargol powder used in this method. Based on an in-house protocol for its synthesis published in 2013, more than 10 batches of protargol powder were produced and subsequently applied in taxonomic studies. During these studies, the protocol for protargol powder synthesis was slightly modified and employed a peptone not original… Show more

“…Furthermore, some species are lost during the preservation protocol with acid Lugol’s ( Gieskes and Kraay, 1983 ; Stoecker et al, 1994 ) or are altered by it, for example by break-up of colonies ( Mukherjee et al, 2014 ), loss of flagella and discoloration ( Karlson et al, 2010 ). Phase contrast and staining agents (e.g., silver staining of ciliates) can help with the identification of colorless or discolored species ( Kim and Jung, 2017 ). The lowest diversity detected with chloroplast 16S rRNA was mainly due to the fact that it only included phototrophs.…”

“…The infraciliature was revealed by protargol impregnation and scanning electron microscopy (SEM). Protargol powder was synthesized using the method of Pan et al ( 2013 ) and Kim and Jung ( 2017 ), and the protargol impregnation technique is based on “procedure A” of Foissner ( 1991 , 2014 ). Cysts were collected from the clone culture or produced by the starvation of large morph specimens.…”
