1 The regulatory e ects of transforming growth factor (TGF)-a on phospholipase A 2 (PLA 2 ) isozymes contributing to prostaglandin generation in rat gastric epithelial RGM1 cells were examined.2 Stimulation with TGF-a for 24 h time-dependently induced prostaglandin E 2 generation with an increase in cyclo-oxygenase-2 protein. The TGF-a-induced prostaglandin E 2 generation was suppressed by NS-398, a cyclo-oxygenase-2 inhibitor. 3 TGF-a stimulated the activity and the protein synthesis of cytosolic PLA 2 (cPLA 2 ). A timedependent increase in cPLA 2 protein occurred in parallel with PGE 2 generation, which was inhibited by methyl arachidonyl¯uorophosphonate (MAFP), a cPLA 2 inhibitor. However, no change in activity of secretory PLA 2 or Ca +2 -independent PLA 2 was observed in the TGF-a-stimulated cells. 4 Stimulation with the Ca 2+ ionophore A23187 for 10 min induced MAFP-sensitive arachidonic acid liberation. Interestingly, preincubation with TGF-a for 24 h diminished A23187-stimulated arachidonic acid liberation despite the increase in cPLA 2 protein.5 Under the conditions, TGF-a was found to increase p11, an endogenous cPLA 2 suppressor, also known as annexin II light chain. The TGF-a-induced increase in p11 was suppressed by tyrphostin AG1478, an inhibitor of tyrosine kinase of epidermal growth factor receptor, which was also found to restore the inhibition by TGF-a of A23187-stimulated arachidonic acid liberation. However, TGF-a did not alter protein levels of annexin II heavy chain. 6 These results suggest that TGF-a stimulates prostaglandin generation through an increase in cPLA 2 , the hydrolytic action of which may be under the control of p11.

The release of arachidonic acid from membrane glycerophospholipids through the action of phospholipases (PLs) is the first step in the biosynthesis of prostaglandins (PGs). In reproductive tissues, the most important PLs are cytosolic PLA 2 (cPLA 2 ) and types IIA and V of the secretory isoform (sPLA 2 ). The aim of this work was to investigate the role of ovarian steroid hormones and oxytocin (OT) in the regulation of rat uterine PLA 2 activity and expression during pregnancy and labor. The activity of sPLA 2 increased near labor, whereas cPLA 2 activity augmented towards the end of gestation. The levels of sPLA 2 IIA and cPLA 2 mRNA showed an increase before labor (P!0.05, day 21), whereas sPLA 2 V mRNA was not regulated during pregnancy. The administration of atosiban (synthetic OT antagonist) together with tamoxifen (antagonist of estrogen receptors) was able to decrease cytosolic and secretory PLA 2 activities, diminish the expression of sPLA 2 IIA and cPLA 2 , as well as decrease PGF 2a production before the onset of labor (P!0.01). The ovarian steroid did not affect PLA 2 during pregnancy. Collectively, these findings indicate that in the rat uterus, both 17b-estradiol and OT could be regulating the activity and the expression of the secretory and the cytosolic isoforms of PLA 2 , thus controlling PGF 2a synthesis prior to the onset of labor. 1 In the zymosan rat air pouch model of inflammation we have assessed the time dependence of phospholipase A2 (PLA2) accumulation in the inflammatory exudates as well as cell migration, myeloperoxidase activity, prostaglandin E2 (PGE2) and leukotriene B4 (LTB4) levels. 2 A significant increase in PLA2 activity was detected in 1,200 g supernatants of exudates 8 h after injection of zymosan into rat air pouch. This event coincided with peaks in cell accumulation (mainly neutrophils) and myeloperoxidase activity in exudates and was preceded by a rise in eicosanoid levels. 3 This enzyme (without further purification) behaved as a secretory type II PLA2 with an optimum pH at 7-8 units, lack of selectivity for arachidonate release and dependence on mM calcium concentrations for maximal activity. 4 The PLA2 inhibitors manoalide and scalaradial inhibited this enzyme activity in vitro in a concentration-dependent manner. Scalaradial also inhibited zymosan stimulated myeloperoxidase release in vitro.5 Injection of the marine PLA2 inhibitor scalaradial together with zymosan into the pouch at doses of 0.5, 1 and 5 umol per pouch resulted in a dose-dependent inhibition of PLA2 activity in exudates collected 8 h later. Myeloperoxidase levels and cell migration were also decreased, while eicosanoid levels were not modified. 6 Colchicine administration to rats prevented infiltration and decreased PLA2 levels in the 8 h zymosan-injected air pouch. 7 These results indicate that during inflammatory response to zymosan in the rat air pouch a secretory PLA2 activity is released into the exudates. The source of this activity is mainly the neutrophil which migrates into the pouch. 8 Scalaradial exerts anti-inflammatory effects in the zymosan air pouch.
