Secretory and cytosolic phospholipase A2 activities and expression are regulated by oxytocin and estradiol during labor

Abstract: The release of arachidonic acid from membrane glycerophospholipids through the action of phospholipases (PLs) is the first step in the biosynthesis of prostaglandins (PGs). In reproductive tissues, the most important PLs are cytosolic PLA 2 (cPLA 2 ) and types IIA and V of the secretory isoform (sPLA 2 ). The aim of this work was to investigate the role of ovarian steroid hormones and oxytocin (OT) in the regulation of rat uterine PLA 2 activity and expression during pregnancy and labor. The activity of sPLA 2… Show more

“…An explanation may be linked to a possible influence of hormonal status on sPLA2-IIa activity. In fact, several studies evidenced that PLA2 activity is modulated by both agonists and antagonists of steroid hormones (e.g., 17b-estradiol, progesterone, and tamoxifen), in breast as well as in other tissues [45][46][47][48]. Another possible mechanism may be related to the significant expression of Annexin II (an endogeneous inhibitor of PLA2 activity) in breast cancer tissues respect to the undetectable levels in normal and hyperplastic breast ductal epithelial cells [49]; the dysregulated balance between sPLA2-IIa activity and its inhibition by annexins has been proposed in the pathogenesis of prostate cancer [50].…”

“…Indeed, in pregnant rats uterine Ca 2? -PLA 2 activity is elevated near gestation in response to increased estradiol concentrations [156]. Estradiol-administration also enhanced the activity of Ca 2?…”
