In this study, the micronucleus (MN) frequency was assessed as a measure of genotoxicity in exfoliated cells of buccal mucosa extracted from 110 petrol pump workers and 100 controls. For each individual, 3,000 exfoliated buccal cells were analyzed. The individuals used in the study were grouped based on their smoking, drinking alcoholic beverages, and tobacco chewing habits. There was a significantly higher frequency of micronucleated cells in the exposed workers to petrol than in the unexposed control population. Smoking and drinking (alcohol) habits, age and length of occupation represent significant factors in terms of increasing the MN frequency measured in the exposed population. This study demonstrates that, using MN assay, it is possible to assess the cytogenetic damage in exposed individuals and that the significant increase in the induction of the MN in the exposed population suggests that the studied individuals may be at a higher risk of developing cancer and therefore monitored for any long term adverse effects of the exposure.

Background:Liquid-based cytology (LBC) can be used for the evaluation of micronucleus (MN) in exfoliated buccal epithelial cells of populations occupationally exposed to potentially carcinogenic agents.Aim:This study was undertaken to evaluate the frequency of MN of exfoliated buccal epithelial cells using LBC preparation in petrol station workers.Materials and Methods:Fifty petrol station workers (cases) and fifty hospital administrative staffs (controls) were recruited and evaluated for MN by May-Grunwald Giemsa, Hematoxylin and Eosin, and Papanicolaou stains using LBC preparation. Statistical analysis was performed with Student's t-test, and P < 0.05 was considered statistically significant.Results:Regardless of staining method used, the mean frequency of MN for cases was significantly higher than that of controls (P < 0.001).Conclusion:The present study concluded that petrol station workers are under risk of significant cytogenetic damage. The MN in exfoliated buccal epithelial cells found to be a useful biomarker of occupational exposure to genotoxic chemicals. LBC can be used for sample preparation to evaluate the frequency of MN in those who are occupationally exposed to potentially carcinogenic agents in view of overall improvement on sample preservation and visualization of cell morphology.

Since 1967, a chemical plant in the town of Chapaevsk ( Samara province, Russia ) has produced large amounts of chlorinated compounds and is suspected to be a major source of local environmental dioxin contamination. Dioxins have been detected in the local air, soil, drinking water, vegetables, and cow's milk. Human exposure to dioxins is suspected as a factor in the deteriorating local public health. In an effort to characterize nonoccupational dioxin exposure among local residents, during the summer of 1998, 24 volunteers were recruited to donate blood and to provide information about their residence, employment, demographics, medical history, and dietary habits. Selected polychlorinated dibenzodioxins, dibenzofurans, and coplanar biphenyls were measured in blood serum samples. The mean concentration of total dioxin World Health Organization toxic equivalents ( WHO -TEQ 98 ) based on polychlorinated dibenzo -paradioxins ( PCDDs ), polychlorinated dibenzofurans ( PCDFs ), and coplanar polychlorinated biphenyls ( PCBs ) was 61.2 ( range 16.4 -168.1 ) pg / g lipid. Subjects living in close proximity to the plant ( less than 5 km ) had significantly higher dioxin levels ( mean WHO -TEQ 98 , 75.7 pg / g lipid ), as compared to subjects living more than 5 km from the plant ( mean WHO -TEQ 98 , 44.1 pg / g lipid ) ( P < 0.04 ). Comparisons of the study results with available published data indicate that average blood dioxin levels were substantially higher in Chapaevsk residents than in nonoccupationally exposed populations of other parts of Russia, Europe, and North America. Chronic exposures of such magnitude may have appreciable adverse effects on public health.
