Cytogenetic impact of sodium chloride stress on root cells of Vigna radiata L. seedlings

Abstract: ObjectivesThe high salinization stress to seedling is the substantial ecological problem in the ongoing era. It negatively influences the growth that retard mitotic division by enhancing aberrations in nuclear chromatin. In the light of these views, the current work was designed to investigate the response of Vigna seedlings root tip cells to the presence of NaCl ions.Materials and methodsNM-92 and NM19-19 seeds were imbibed separately in distilled water for 24 h and allowed to grow into 0, 50, 150, 250, and 3… Show more

“…Twenty healthy seeds were imbibed separately in pretreatment solution of 0.01 mM Cd, Ni, Hg, Pb, Cu and distilled water (control) for 24 h, allowed to germinate in Petri dishes with distilled water for 24 h. Later treated with five respective metals of 0.1 mM and 0.5 mM concentration for 5 h. Root tips were collected for cytological analysis. Root tips were washed with distilled water thrice and fixed in Farmer s fixative solution (3 : 1 absolute alcohol: glacial acetic acid) for 24 h, later stained in 1.8% aceto-orcein according to Fiskesjo (1997) with minor modifications (Farheen and Mansoor 2019). Stained was prepared by mixing 1.8 g of aceto-orcein in 100 mL of 45% glacial acetic acid followed by boiling in a water bath for 7 h on a hot plate (M6).…”

“…While food colour additives were group-2 that was first placed in the distilled water filled beaker until the adventitious root of onion bulbs became 1-2 cm long. Then onion bulbs were transferred into the separate beakers having different colour additives for 36 h. After colour additives treatment, onion root tips were cut and preserved in "Farmer Fixative" for 24 h (Farheen and Mansoor, 2019). The onion root cells were hydrolyzed in the 1N hydrochloric acid solution for 5 minutes at 60 °C then stained in 1.8% aceto-orcein for 24 h (Farheen and Mansoor, 2019).…”
