Cytogenetic and molecular evidence suggest multiple origins and geographical parthenogenesis in Nothoscordum gracile (Alliaceae)

Abstract: The 2n = 19 karyotype was probably formed by a centric fusion event occurring in N. nudicaule and later transmitted to tetraploid cytotypes of N. macrostemon. Diploids of N. nudicaule and N. macrostemon appeared as consistent recently diverged species, whereas tetraploid apomicts seem to constitute an assemblage of polyploid hybrids originating from multiple independent hybridization events between them, part of which are morphologically recognizable as N. gracile.

“…-Root tips from specimen L.P Felix 10803 were pretreated with 0.002 M of 8-hydroxyquinoline for 24 h at 10 ºC and fixed in Carnoy's solution. CMA/DAPI banding and FISH procedures were performed according to Souza et al (2012). Fixed root tips were washed in distilled water and digested in a 2% (w/v) cellulase (Onozuka)/20% (v/v) pectinase (Sigma) solution at 37 ºC for 120 min and macerated in a drop of 45% acetic acid; the coverslip was later removed in liquid nitrogen.…”

“…Marginal distributions of species are characterized by greater heteromorphism and wide spectra of chromosomal variations including supernumerary chromosomes, heterozygosity for translocations, polyploidy, and variability of the chromosomal positions and/or numbers of 45S and 5S ribosomal DNA (rDNA) sites (Belyayev and Raskina 2013;Souza et al 2012). These changes in karyotype structure may be regarded as elementary events of microevolution related to intense pressure from environmental factors (e.g., edaphic and climatic conditions) and can occur in parallel with changes in plant morphology and physiology (Martienssen 2008;Belyayev and Raskina 2013).…”

“…Additionally, species of the same genus can differ from each other in terms of the number and position of their heterochromatic bands or rDNA sites (Souza et al 2010(Souza et al , 2012. Therefore, a more detailed comparative cytogenetic approach could contribute to elucidate the relationships between the species of Leucocoryne with three and six stamens.…”

“…Even at different blocking DNA concentrations (1X, 25X, 50X, and 75X), staining remained the same. The fact that this complex is very new may have been the reason that GISH was unable to distinguish the genomes belonging to the Inodorum complex, making it possible to produce viable hybrids between N. nudicaule and N. macrostemon (Souza et al, 2012).…”
