Background There is a morphological overlap among renal epithelial tumors, particularly chromophobe renal cell carcinoma (CHRCC), clear cell renal cell carcinoma (CCRCC), renal oncocytoma (RO), and papillary renal cell carcinoma (PRCC). Discriminating between these tumors is important but sometimes challenging. This study is aimed at evaluating the clinical usefulness of the combined immunochemistry for the “three 7” markers (CK7, CD117, and Claudin-7) to distinguish chromophobe renal cell carcinoma from these mimics. Methods Immunochemical staining for CK7, CD117, and Claudin-7 was performed in 68 CHRCCs, 199 CCRCCs, 32 ROs, and 30 PRCCs. Fluorescence in situ hybridization (FISH) was performed in some cases to exclude CCRCC and PRCC. The sensitivity (SE) and specificity (SP) for CHRCC as well as the immunoreactivity of each marker and their combinations were statistically evaluated. Results High positive rates for CK7 (94%), CD117 (87%), Claudin-7 (94%), and their combinations (CK7+CD117, 79%; CK7+Claudin-7, 88%; CD117+Claudin-7, 82%; CK7+CD117+Claudin-7, 76%) were observed in CHRCC compared to those in CCRCC, RO, and PRCC, with increasingly higher SP when combinations of the “three 7” markers were applied (CK7, 0.80; CD117, 0.82; Claudin-7, 0.78; CK7+CD117, 0.95; CK7+Claudin-7, 0.97; CD117+Claudin-7, 0.97; CK7+CD117+Claudin-7, 1). Conclusion CK7, CD117, and Claudin-7 are frequently expressed in CHRCC with high specificity. We recommend the routine use of these 3 markers as a routine panel when making a differential diagnosis of CHRCC and excluding other mimics.

Each histological variant of renal cell tumors has a unique color. The yellowish color of clear cell renal cell carcinoma (CCRCC) is explained by the presence of intracytoplasmic lipid and glycogen accumulation. Color changes in CCRCC are correlated with clinicopathological and metabolic changes, as well as biological behavior. We analyzed and compared the clinical, histopathological, and immunohistochemical features and gene expression profiles, in lipid metabolism of yellowish-colored ChRCC (ChRCC-Y), non-yellowish-colored ChRCC (ChRCC-N), and CCRCC. Of 14 ChRCCs, we retrieved 6 ChRCC-Ys. Patients with ChRCC-Y are younger than those with ChRCC-N, and the tumor is not predominant in males. ChRCC-Ys are smaller than ChRRC-Ns. Three ChRCC-Ys exhibited individual discrete tubule formation. No ChRCC-Ns exhibited individual discrete tubule formation. Two of 6 ChRCC-Ys showed relatively diffuse adipophilin positivity. No ChRCC-Ns demonstrated diffuse positivity for adipophilin. The expression of SCD, FDFT1, and E2F1 showed a tendency to be lower in ChRCC-Y than in ChRCC-N. The expression of PDGFB showed a tendency to be higher in ChRCC-Y than in ChRCC-N. This study demonstrated ChRCC-Y did not indicate an increase in lipid and cholesterol metabolism and that ChRCC-Y did not have the common molecular alteration of CCRCC. The absence of such metabolic acceleration in ChRCC-Y might support the biological indolent behavior. Furthermore, we revealed that macroscopic color changes might be correlated with various clinicopathological features and immunohistochemical and molecular changes from different perspectives. We believe further characterization of RCC, including tumor heterogeneity, is needed to improve the management of patients with RCC.

Background: Systemic lupus erythematosus (SLE) is a chronic immune connective tissue disease, which is common in women of childbearing age and easy to cause multiple organ inflammatory injury. The occurrence of prostate cancer is the result of multiple factors and genes, but we have little understanding of the mechanism involved. In this study, we deeply explored and analyzed the existing gene data in GEO database in order to find the key genes and new therapeutic targets of SLE.Results: The expression profile dataset of GDS4185, GDS4888, GDS4889 and GDS4890 containing 99 specimens, 42 cases of SLE patients and 57 cases of normal volunteers, were downloaded from the Gene Expression Omnibus (GEO) website. The differentially expressed genes (DEGs) in different tissues was analyzed by statistical hypothesis T test. The gene ontology (GO) enrichment analysis was carried out by the DAVID online tool. KEGG pathway annotation of DEGs was carried out by the KOBAS online computing database. The protein–protein interaction (PPI) networks of the DEGs were built from the STRING website and Cytoscape software. A total of 839 DEGs were calculated from the four GEO datasets. The GO and KEGG analysis indicated that the functions of DEGs mostly participated in the Osteoclast differentiation, HTLV-I infection, Measles, FoxO signaling pathway, Herpes simplex infection, Primary immunodeficiency, Jak-STAT signaling pathway. The following 14 closely related genes, HERC5, TP53, CDC20, GNB2, GNB4, PPP2R1A, GNAI2, PMCH, SOCS3, HERC6, STAT1, SOCS1, ISG15, IFIT3, were key nodes from the PPI network. These genes may have synergistic or indirect interactions with each other in the process of biological metabolism inducing the pathogenesis of SLE.Conclusion: Mining geo database has great scientific research value. In the future, scientific research must fully excavate a variety of database analysis methods. In this study, the screened candidate genes provide effective theoretical basis for the diagnosis, treatment, expected evaluation and related laboratory research of SLE, which are worthy of further experimental verification.
