Cytochrome oxidase response to cochlea removal in chicken auditory brainstem neurons

Abstract: Changes in cytochrome oxidase (CO) activity were studied in the chick brainstem auditory nuclei, n. magnocellularis (NM) and n. laminaris (NL), following unilateral cochlea removal. Chickens aged 10 days or 56 weeks underwent unilateral cochlea removal. Following survival periods of 30 minutes to 14 days for the 10-day-old birds and 6 hours or 14 days for the 56-week-old birds, the animals were perfused with paraformaldehyde/glutaraldehyde fixative. Cryostat sections of the brainstem were then prepared for CO … Show more

“…The quantitative and statistical procedures are similar to those described in detail in earlier publications (Born and Rubel, 1988;Hyde and Durham, 1990;Garden et al, 1994). Absolute ODs of labeled neurons vary among brain sections and among the brains of different animals because of a host of factors, such as perfusion, rinsing, age of antibody, and variability of the chemical reaction that produces the colored precipitate.…”

“…Typically the final reaction product of CO histo chemistry is qualitatively described, although, oth ers have utilized optical densitometry for quantifi cation (Hovda et al, 1987;Darriet et al, 1986;Kageyama and Meyer, 1988;Hyde and Durham, 1990;Wong-Riley and Kageyama, 1986). This method of quantification appears to be justified as long as the factors which can influence the density of the reaction in vitro are controlled for (Silverman and Tootell, 1987).…”

“…Brains were cut in 20-m-thick coronal sections on a cryostat, and alternate sections were thaw mounted on slides to form two series. One series was stained for cytochrome oxidase to show brainstem cytoarchitecture (Hyde and Durham,, 1990). The other series was apposed to Kodak Biomax MR X-Ray film to obtain images of 2DG distribution within the brain.…”
