Cyclosporin a in the Treatment of Severe Behçet’s Uveitis

Abstract: Twelve patients with active Behçet's uveitis with marked deterioration of visual acuity in at least one eye were treated with cyclosporin A (CyA). An initial improvement in the severity of ocular inflammation and systemic features occurred in all cases and persisted until the dose was reduced or the drug withdrawn when a rapid recurrence of symptoms was noted. The visual acuity also initially improved in ten patients and this was maintained in seven cases until the dose of CyA was reduced. At this time, acuity… Show more

“…It has been shown in so many studies that flare and cells of vitreous decrease within the first few weeks of therapy, while retinal vasculitis needs a longer period of time for improvement (Mü ftü oglu et al 1987;Binder et al 1987). Withcup et al (1994) have shown that, in comparison to CSA alone, combined CSA with steroid treatment has been successfully effective in improvement of visual acuity within the first three months of treatment, but there were no significant differences in overall visual acuity of two groups in a long term period.…”

“…CsA is a fungal cyclic undecapeptide that has been shown to be effective both in organ transplantation [15] and in many autoimmunue disorders such as Behcet's syndrome [2,4], psoriasis [8] and rheumatoid arthritis [6]. CsA like FK506 acts by inhibiting IL2 and IL2R production and the early stages of Tlymphocyte activation.…”

“…In this open clinical trial, intermittent, low-dose cyclosporine was found to abrogate the intraocular inflammation. Improvement in clinical findings may occur within three to five days in experimental studies and within one to two weeks in clinical trials [21][22][23]. In this study, we found out that exacerbations were frequent in patients with Behcet's disease.…”
