Cyclodextrin-Based Molecular Accessories for Drug Discovery and Drug Delivery

Abstract: Recently, active pharmaceutical ingredients (APIs) have been dramatically expanding from low-molecular weight drugs to peptides, proteins, antibodies, genes, oligonucleotides, cells, and machines. Therefore to develop pharmaceutical technologies and drug delivery systems (DDS) for these APIs, advanced molecules and novel concepts are needed. In this review, we introduce cyclodextrin-based supramolecular accessories such as molecular necklace, molecular earring, and intelligent molecular necklace, and describe … Show more

“…Cyclodextrins are still regarded as "novel" excipients, active ingredients, drug delivery vehicles and anti-aggregation agents (Conceição et al 2018). New formulations continue to be reported, generating new interests in medicine and biomedicine (Higashi et al 2018;Higashi 2019;Menezes et al 2019;Pawar and Shende 2019). The 18 ongoing clinical trials in 2019 give the promise of new marketed formulations (Cyclodextrin News, CycloLab Ltd., Hungary).…”

“…Cyclodextrins (CD, Figure 2), cyclic oligosaccharides derived from starch, are the most popular and developed drug delivery systems [22,23]. CDs have a hydrophilic shell and hydrophobic core suitable for the inclusion of lipophilic “guest” molecules of appropriate size.…”

“…Recent findings have also suggested that polypseudorotaxane hydrogels consisting of α- or γ-CD and PEG (mw 20,000 Da) markedly improved stabilities of human IgG against heating and shaking ( Higashi et al, 2015 ), suggesting them as stabilizers for antibody drugs without changing their pharmacokinetics ( Fig. 4 B) ( Higashi, 2019 ).
