Vascular smooth muscle cell (VSMC) hyperproliferation is the main pathological process in various cardiovascular diseases, such as vascular restenosis. This process may be repressed by RING finger protein 10 (RNF10) in metabolic syndrome (MetS) rats. The aim of this study is to evaluate the inhibitory effects and molecular mechanisms of RNF10 on VSMC hyperproliferation. Neointimal hyperplasia in MetS and high‐glucose‐induced VSMC hyperproliferation were measured after infection with adenoviruses encoding RNF10 (Ad‐RNF10), short hairpin RNF10 (Ad‐shRNF10), or green fluorescent protein (Ad‐GFP). In vivo and in vitro, we found that overexpression of RNF10 significantly affected neointima formation and VSMC proliferation, and displayed further inhibitory activity by promoting mesenchyme homeobox 2 (Meox2) and suppressing activating protein 1 (AP‐1). In contrast, Ad‐shRNF10 had an opposite effect on neointimal hyperplasia and VSMC hyperproliferation in vivo and in vitro. Our study indicated that RNF10 inhibited the hyperproliferation with the activities of Meox2 and AP‐1 proteins. RNF10 may be a next drug target for treating vascular restenosis and other related cardiovascular diseases. © 2018 IUBMB Life, 71(5):632–642, 2019

Anthocyanins are a family of natural pigments classified into the group of flavonoids, considered to be responsible for the color and taste of many fruits and vegetables, i.e. berries. Anthocyanins are common components of the human diet. Besides their interest as colorant because of their coloring properties, the study of anthocyanin compounds stems from their wide applicability in the prevention and even in the treatment of various human diseases. However, various aspects of the pharmacological roles of anthocyanins remain in the dark, having still several obstacles to the development of robust diets or prescribing lines on consumption of anthocyanins. The chemical structure of anthocyanins determines in large measure its capacity and efficacy as an antioxidant agent. In this study, the following aspects are reviewed: the antioxidant effect of anthocyanin pigments; the oxidative stress, the bioavailability after intake and biological aspects of anthocyanins, the method for measuring the antioxidant activity of anthocyanins, the relationship between structure and activity; and the influence of the anthocyanins in the antioxidant activity of wines. Finally an overview of some potential uses in food industry is attempted mainly focusing in the anthocyanin encapsulation topic. Attention has been paid to the more recent publications in the field.

Berry consumption shows antiobesity and antidiabetic effects in animals and humans.Anthocyanin or berry extracts increase insulin sensitivity, reduce lipid droplet formation, and cause a white-to-brite phenotypic switch of adipocytes in vitro. The objective of this study was to investigate effects of cyanidin-3-glucoside (C3G) or blackberry extract (BBE) on cellular respiration of a fatty acid substrate in differentiated adipocytes. In separate experiments, 3T3-L1 adipocytes were treated with 0, 150 nM, or 50 μM purified C3G -or-0, 150 nM, 25 μM, or 50 μM C3G-equivalents from a BBE. Baseline, state-4, state-3, and uncoupled respiration states were created experimentally and measured by high-resolution respirometry. Respiration rate within each state was corrected for mitochondrial content determined by PCR. Mitochondrial content was significantly reduced with high-dose C3G and the two high-dose BBE. The high-dose C3G caused a significant increase in respiration rate for baseline, state-4, state-3, and uncoupled states compared to control and physiologic dose C3G. The intermediate-high dose (25 μM) BBE also significantly increased respiration rate for the same states compared to control and other treatment levels of BBE; this effect was not observed at the high-dose (50 μM) BBE. Administration of high-dose purified C3G or BBE decreased mitochondrial content but enhanced cellular respiration on a fatty acid substrate in differentiated 3T3-L1 adipocytes across several mitochondrial states. We did not observe a phenotypic switch to brite adipocytes like others, but our empirical findings in cellular respiration on a fatty acid substrate support the antiobesity effects associated with anthocyanins and berries.
