Current High-Throughput Approaches of Screening Modulatory Effects of Xenobiotics on Cytochrome P450 (CYP) Enzymes

Abstract: Cytochrome P450 (CYP) is a critical drug-metabolizing enzyme superfamily. Modulation of CYP enzyme activities has the potential to cause drug–drug/herb interactions. Drug–drug/herb interactions can lead to serious adverse drug reactions (ADRs) or drug failures. Therefore, there is a need to examine the modulatory effects of new drug entities or herbal preparations on a wide range of CYP isoforms. The classic method of quantifying CYP enzyme activities is based on high-performance liquid chromatography (HPLC), … Show more

“…S. cerevisiae was the first system and one of the top choices for heterologously expressing eukaryotic P450s, and various P450s have been functionally expressed in S. cerevisiae (van Leeuwen et al, 2012; Oeda et al, 1985; Ung et al, 2018). Urban et al (1994) successfully expressed Helianthus tuberosus P450 (CYP73, cinnamate 4‐hydroxylase) in S. cerevisiae for the production of p‐coumarate.…”

“…However, herb-drug interactions (HDI) might occur during the process of clinical application. The important metabolic enzyme, cytochrome P450, plays a vital role in drug metabolism [35]. CYP450 is a superfamily of enzymes, and the majority of CYPs involved in drug metabolism include CYP3A4/5, CYP2D6, CYP1A2, CYP2B6, CYP2C19, CYP2A6, and CYP2J2.…”

“…The fluorescence-based approach for screening has several advantages: it is faster and much less expensive than LC-MS methods. Data can be generated quickly without compromising data quality [ 10 , 36 , 71 , 72 ].…”
