Curcumin Enhances Cytotoxic Effects of Bortezomib in Human Multiple Myeloma H929 Cells: Potential Roles of NF-κB/JNK

Abstract: Combined curcumin and PS-341 treatment has been reported to enhance cytotoxicity and minimize adverse effects through ERK and p38MAPK mechanisms in human multiple myeloma cells. However, whether JNK plays similar role in this process remains unclear. In the present study, we found combined treatment altered NF-κB p65 expressions and distributions in multiple myeloma H929 cells. Western blot analysis showed combined treatment inactivated NF-κB while activated JNK signaling. Pre-treatment with JNK inhibitor SP60… Show more

“…These findings are similar to those of previous studies with myeloma cell lines (U266 and H929) using curcumin and a less potent immunomodulatory drug, i.e. thalidomide [10] and bortezomib [11] respectively. The H929 cell line was chosen for this study because it harbours the t(4; 14) translocation, whereas the U266 cell line carries a t(11; 14) translocation [18] .…”

“…Figure 2 depicts the hierarchical cluster analysis of the obtained gene expression patterns. Previous studies highlighted that treatment with curcumin and bortezomib activated JNK signaling in multiple myeloma cells [ 33 ]. In agreement with that, we observed increased expression of JUN (jun protooncogene), a target of JNK (MAPK8, mitogen-activated protein kinase 8), following both curcumin and bortezomib treatment, and this overexpression was enhanced by their combined use.…”

“…It has attracted much interest due to its low toxicity, proven pharmacological safety, and favorable biological activity, which includes lowering of blood fat as well as anti-inflammatory and antitumor effects. Its antitumor effects, both alone and combined with other chemotherapeutics, are widely supported by recent studies, and one of these has shown that curcumin can inhibit the proliferation of malignant plasma cells (myeloma cells) and induce apoptosis [7]. Therefore, we hypothesized that curcumin and bortezomib in combination could have a synergistic effect on myeloma cells.…”
