Curcumin derivatives as photosensitizers in photodynamic therapy: photophysical properties and in vitro studies with prostate cancer cells

Abstract:

Increased absorption maximum for the dimethylaminocurcumin with strong PD effect on prostate cancer cells; Intense PD effect of curcumin III on prostate cancer cells; Exciting biphasic PD response by curcumin I and cinnamaldehyde-derived curcumin.

“…Based on this hypothesis, the antifungal effect of bisdemethoxycurcumin might be lower than that of demethoxycurcumin, but in our study, a dramatic reduction in Candida albicans cells was observed as caused by the bisdemethoxycurcumin-containing test photosensitizer, and thus we hypothesized that the effect of ROS itself plays a more major role than the fungal cell adhesion ability of photosensitizers in Candidiasis suppression. This was also found in the study of Kazantzis et al [ 29 ], showing evidence that the lack of the methoxy-aromatic substitution in bisdemethoxycurcumin favors a high degree of free radical production.…”

“…Synthetic derivatives of curcumin, cinnamaldehyde and dimethylamino derivative, were reported to improve sensitization of LNCaP cells for photodynamic therapy at concentration of 3 μM. Curcuminoid dimethylamino derivative reduced cell survivability in a dose-dependent manner ( Kazantzis et al, 2020 ). Performing curcumin derivatives and photodynamic therapy together on other PCa cell lines and in animal models might give a novel method for curbing this disease.…”

“…A highly conjugated chemical structure results in visible light spectrum absorption, as well as mediation of reactive oxygen species generation upon irradiation. These facts mean that CUR and its derivatives are also considered as potential photosensitizers in photodynamic therapy (PDT) against tumors and infectious diseases [25,26]. Another property that impacts the biological activity of curcuminoids is their tendency to form strong metal complexes.…”
