“…The proportion of binucleate trophoblast giant cells among other cotyledonary cells was moderately higher than that found by others (Landim et al, 2007;Vanselow et al, 2008), showing a relative proportion of about 30-40% after percoll gradient centrifugation and ablation of fibroblast cells. Perhaps this might be due to different isolation periods of cotyledonary and different time of disaggregation.…”

“…Since AP-2 proteins have a featured helix-span-helix motif at the carboxyl terminus and a central basic region, which can form homo-or hetero-dimeric complexes with some specific novel group of coactivators to bind to a particular promoter (Eckert et al, 2005), by this way they can modulate transactivation capability or DNA-binding affinity of the other AP-2 family members, thus further experiments are required to identify the specific properties of each complex when the CSH1, PRP1 and PAG1 gene are in the inactive and active states. In addition, AP-2 proteins can be viewed as gatekeepers controlling the balance between proliferation and differentiation during embryogenesis (Hilger-Eversheim et al, 2000;Landim et al, 2007), therefore, the signaling pathways responsible for induction of AP-2 genes are currently under investigation, these information will provide a better understanding of placentation and embryogenesis.…”

“…27,28 We used serum deprivation and low oxygen tension as 2 sources of stress since each is known to induce enhanced apoptosis in trophoblasts. [29][30][31][32] Trophoblasts cultured in vehicle control or ET-1 were exposed to serum-free medium in 20% oxygen for a total of 16 hours, or 24 hours prior to harvest at 72 hours, or to an environment with an oxygen concentration of 8%, which resembles villous oxygen tension in vivo after the first trimester 33 or <1% oxygen, in addition to standard 20% oxygen. The medium was equilibrated with each gas mixture 30 minutes prior to use.…”

“…Com passar dos anos e desenvolvimento de novas tecnologias, pesquisadores conseguiram estabelecer um protocolo efetivo para cultivo de CGT bovina pelo período de 90 dias. Durante este período as CGTB eram isoladas das demais CGT onde permaneciam vivas até mesmo na presença do substrato colágeno (LANDIM JR. et al, 2007). Este achado pode ser explicado pelo fato da cultura utilizada ser composta de 90% de CGT e 10% de fibroblastos.…”
