Cultivation of Denitrifying Bacteria: Optimization of Isolation Conditions and Diversity Study

Abstract: An evolutionary algorithm was applied to study the complex interactions between medium parameters and their effects on the isolation of denitrifying bacteria, both in number and in diversity. Growth media with a pH of 7 and a nitrogen concentration of 3 mM, supplemented with 1 ml of vitamin solution but not with sodium chloride or riboflavin, were the most successful for the isolation of denitrifiers from activated sludge. The use of ethanol or succinate as a carbon source and a molar C/N ratio of 2.5, 20, or … Show more

“…Most Probable Number (MPN) determination) in four different anaerobic cultivation media: (1) LB medium (Bertani 1951) for detection of heterotrophic anaerobic microorganisms; (2) R2A medium (Reasoner and Geldreich 1985) for slow-growing microbes; (3) Medium 63 (Deutsche Sammlung von Mikroorganismen und Zellkulturen) for the cultivation of sulfate-reducing prokaryotes (SRP); (4) N43 medium (Heylen et al 2006) to detect nitrate-reducing prokaryotes. All media contained activated resazurin (1 mg L -1 ) as redox indicator to verify anoxicity.…”

“…Thauera sp. could produce lots of extracellular polymeric substances which contributed to the granulation process (Heylen et al, 2006). Chloroflexi bacteria were facultative anaerobes and were important for granulation (Chen et al, 2013).…”

“…Within the dominant gammaproteobacterial nitrate reducers, seven isolates were retrieved on the defined isolation medium G3M12 -with ethanol as carbon source and nitrate as nitrogen source (Heylen et al, 2006) -that could be assigned to Stenotrophomonas based on partial 16S rRNA gene sequences. Three of these isolates, R-32746, R-32768 T and R-32729 T , could possibly represent novel species based on partial 16S rRNA gene sequence similarities and were analysed further in a polyphasic study.…”

“…The temperature range (at 4, 15, 28, 37 and 52 u C), pH range (4.5-10.5 at 28 u C) and salinity range (0.5-5 % w/v, at 28 u C) for growth were recorded after incubation for 72 h on TSA. The ability to reduce nitrate was tested, as described by Smibert & Krieg (1994), after growth for 2 weeks on tryptone soy broth (TSB; Oxoid) supplemented with 10 mM potassium nitrate at 37 u C and on liquid G3M11 medium containing 18 mM potassium nitrate and 22.5 mM ethanol (Heylen et al, 2006) at 20 uC -the solid variant of the latter was used as isolation medium for the Stenotrophomonas strains. For all strains, these results were in agreement with the nitrate reduction test in the API 20NE system.…”
