Cu(II) Complexes of Isoniazid Schiff Bases: DNA/BSA Binding and Cytotoxicity Studies on A549 Cell Line

Abstract: A series of isonicotinoyl hydrazones have been synthesized via template method and were complexed to Cu(II). The ligands are coordinated to Cu(II) ion through the enolic oxygen and azomethine nitrogen resulting in a square planar geometry. The CT-DNA and bovine serum albumin binding propensities of the compounds were determined spectrophotometrically, the results of which indicate good binding propensity of complexes to DNA and BSA with high binding constant values. Furthermore, the compounds have been investi… Show more

“…This was observed in the Co and Ni complexes which showed C=O stretching frequencies at 1649 and 1651 cm -1 as against the C=O stretching vibration. The disappearance of the C=O band at 1669 cm -1 and subsequent appearance of the enolic C-O stretching band at 1055, 1059, 1063 cm -1 for the Mn(II), Cu(II) and Zn(II), respectively, indicates enolization thus isoniazid coordinated to the metals via the enol oxygen [16]. There was an appearance of new bands at 627-637 and 415-419 cm -1 which are due to M-O and M-N bands, respectively [10].…”

“…The k q values are higher than those characterizing diverse kinds of quenchers, pointing towards the existence of a static quenching mechanism [42]. Using the Scatchard equation [43] (Figure 11) and n is given by the ratio of the y intercept to the slope. It is obvious (Table 1) that the coordination of Co(III) complexes results in a decreased K value for BSA with complex 1 exhibiting the highest K value among the other complexes.…”

“…The CT-DNA and bovine serum albumin binding ability of a series of isonicotinoyl hydrazones and their Cu(II) complexes indicated good binding to DNA and serum albumins (BSA). The compounds have cytotoxicity on A549 human lung cancer cell [4]. Moreover, series of pyridazinyl hydrazones inhibit tyrosine hydroxylase and dopamine hydroxylase in vivo and in vitro [5].…”
