Cryptic speciation in Pelobates fuscus (Anura, Pelobatidae): evidence from DNA flow cytometry

Abstract: The amount of nuclear DNA in 173 specimens of Pelobates f. fuscus from 34 localities in Russia, Ukraine, Moldavia and Latvia was determined by DNA ow cytometry. Two distinct groups with different genome sizes were identi ed. The ranges of the genome size variation in the two groups did not overlap. Geographically, these groups with smaller or larger genome size are distributed in the west and in the east of eastern Europe, respectivel y.

“…Moreover, only one animal in the sample showed a somewhat strong preference in its (right) forelimb use. This result is in well correspondence to previ-ous findings in other populations (Malashichev, 2002), all belonging to the same "Eastern" type of P. fuscus (Borkin, Litvinchuk, Rosanov, and Milto, 2001). Similarly, in both species of true frogs (Ranidae) examined, the more terrestrial Grass frog, R. temporaria (Fig.…”

“…In total, 47 individuals were tested for nuclear DNA analysis (8 P. ridibundus, 4 P. lessonae and 22 P. esculentus). The technique is described in details previously (Borkin et al, 2001). The obtained results confirm the water frogs identity, P. ridibundus has 16.2 ± 0.1 pg of nuclear DNA (min-max: 16.1-16.3) P. lessonae -13.9 ± 0.1 pg (13.8-14.1) and P. esculentus -15.2 ± 0.1 pg (14.9-15.6).…”

“…Blood of grass frogs, Rana temporaria Linnaeus, 1758 were used as a reference standard. The details of our technique have been previously published (Borkin et al 2001a;Litvinchuk et al 2004). Voucher specimen locations are given in Appendix S1.…”
