Cryopreservation of pollen from two rose cultivars

Abstract: An investigation was undertaken on the storage characteristics of pollen collected from two English rose cultivars. A rapid decline in viability was observed in pollen stored at + 4 ° C and-20 ° C, whereas the viability of pollen, stored at ultra-low temperature (-196 ° C), remained constant. Cryopreserved pollen was shown to retain its ability for fertilisation. The effects of the stage of flower development and anther dehiscence were assessed on both pre-and post-cryopreservation viabilities. Successful long… Show more

“…The media were prepared with 0.015 g boric acid (Jicinska et al 1976;Pearson and Harney 1984;Gudin et al 1991a, b;Marchant et al 1993) dissolved in 200 mL water. Saccharose was added and the volume was adjusted to 300 mL.…”

“…The success or failure of germination depends on such factors as the period of the year during which the pollen is collected, the stage of development of the flower from which the pollen is collected, and the drying and storage techniques (Marchant et al 1993). Depending on the study, it is recommended that the pollen be collected when the sepals are open (Pearson and Harney 1984), when the outside petals begin to open (Marchant et al 1993) or when the flowers are fully open (Rajasekharan and Ganeshan 1994).…”

“…So far cryopreservation procedures have been published for 21 species and varieties of roses. The first processes of germplasm cryopreservation were carried out on pollen (Marchant et al, 1993;Rajasekharan and Ganeshan, 1994). In recent years, buds…”

“…However, published reports on this aspect are restricted to a few ornamental crops. Hughes & Lee (1991) successfully preserved Clianthus pollen at -1800 C. Marchant et al (1993) cryopreserved pollen of two English rose cultivars for 8 weeks. Among bulbous ornamentals, long-term pollen storage of Narcissus in liquid nitrogen is reported by Bowes (1990).…”
