CRTH2 mediates the activation of human Th2 cells in response to PGD2 released from IgE/anti‐IgE treated nasal polyp tissue

Abstract: These data suggest that in immunologically activated nasal polyp tissue, PGD(2) produced by mast cells promotes the migration of Th2 cells through a CRTH2 dependent mechanism.

“…The role of CRTH2 receptor in mediating chemotaxis of CD4+ Th2 lymphocytes in response to mast cell released substances has been observed [86]. Further, the migration of Th2 cells through a CRTH2 dependent mechanism via mast cells released mediators in nasal polyp tissue has also been reported [87]. More studies needed in this direction to explore the effect of lymphocyte and VIP interaction on the immune system.…”

“…Mast cells are one of the main sources of pro-inflammatory eicosanoids, such as prostaglandin D 2 (PGD 2 ) and cysteinyl leukotrienes (cysLTs), which can affect allergic type-2 responses, activating cells such as Th2, ILC2, eosinophils, and basophils and also acting in autocrine or paracrine fashion on mast cells themselves. 1 It has been established that cysLTs can potently regulate mast cells functions through cysteinyl leukotriene type 1 receptor (CysLT 1 ) but it is still unknown whether PGD 2 can induce activation of human mast cells. 2,3 Conflicting reports have been published showing positive or negative staining for PGD 2 receptors in human tissue mast cells using immunochemistry.…”

“…Activation of Th2 cells by PGD 2 is thought to occur predominantly through DP2/CRTH2 with concomitant increase in the production of cytokines and pro-inflammatory proteins (106, 113115). PGD 2 binding to this receptor is also very important for CD4+ T cell trafficking and motility (116, 117).…”
