CRISPR/Cas9-Mediated Knockout Reveals the Involvement of CYP304F1 in β-Cypermethrin and Chlorpyrifos Resistance in Spodoptera litura

Abstract: Functions of insect CYP2 clan P450s in insecticide resistance are relatively less reported. In Spodoptera litura, a gene from the CYP2 clan (CYP304F1) was validated to be up-regulated significantly in a pyrethroid- and organophosphate-resistant population (QJ) than a susceptible population by RNA-Seq and qRT-PCR. Spatial–temporal expression indicated the high expression of CYP304F1 in the fourth, fifth, and sixth instar larvae and the metabolism-related tissue fat body and malpighian tubules. CYP304F1 was knoc… Show more

“…The overexpression of CYP321B1 plays roles in β‐cypermethrin and cyhalothrin resistance validated by RNAi (Xu et al, 2020). The relative expression of SlGSTe8 was found to be significantly higher in the QJ population than the GX population by RNA‐Seq in our previous study (Xu et al, 2022) and validated by qRT‐PCR in this study. S. litura has relatively large amounts of GSTs genes among most insect species (Hu et al, 2022).…”

“…In Bombyx mori, BmGSTO was induced by diazinon, permethrin, imidacloprid, E. coli and UV-B and might be involved in protecting cells against oxidative stress (Yamamoto , 2011). In Nilaparvata lugens, a notorious insect with serious insecticide resistance, NlGSTd2 was reported to confer resistance to pyrethroid insecticide as an antioxidant agent (Vontas et al, 2001(Vontas et al, , 2002; NlGSTs1 and NlGSTs2 conferred resistance to chlorpyrifos, fipronil, etofenprox, imidacloprid and even the secondary metabolites from host plant through antioxidation function (Zhang et al, 2022). In this study, the recombinant protein SlGSTE8…”

“…This outcome is particularly encouraging as S. litura is among the species of insects which has developed resistance towards CPF. [18] There are several reports in literature regarding the development of bio-based safe pesticides but their synthesis is not convenient (multistep, drastic reaction conditions). [19,20] In contrast, PBI-SAH building block is easy to prepare and is highly fluorescent in aqueous media.…”

“…The present investigation also describes the high efficiency of PBI‐SAH assemblies to disturb the life cycle of Spodoptera litura ( S. litura ) and to exhibit 100 % insecticidal activity after 19 days. This outcome is particularly encouraging as S. litura is among the species of insects which has developed resistance towards CPF [18] . There are several reports in literature regarding the development of bio‐based safe pesticides but their synthesis is not convenient (multistep, drastic reaction conditions) [19,20] .…”
