Pfaffia glomerata (Spreng.) Pedersen is a medicinal species of great interest because it produces the phytoecdysteroid 20-hydroxyecdysone (20E). Generally, because of atypical growing conditions, in vitro propagated plants function less efficiently as autotrophs and have poorly developed morphological structures. This study analyzed the autotrophic potential of P. glomerata propagated in vitro and evaluated the influence that this has on 20E biosynthesis. Physiological and structural parameters of plants subjected to heterotrophic, photomixotrophic and photoautotrophic growth conditions were evaluated. Levels of 20E were measured by HPLC. Plants were acclimatized in a mixture of soil, sand and substrate, in a greenhouse. Conditions that provided higher carbon input led to an increase in plant growth, and the presence of sucrose was critical, in closure systems without a gas permeable membrane, for normal anatomical development of the micropropagated plants. The absence of sucrose increased photosynthesis and conditions that enhanced photoautotrophy induced greater levels of 20E. The increase of 20E levels by the photoautotrophic system offers new prospects for increasing the commercial production of this species, and for studies that could elucidate the biosynthetic pathway of phytoecdysteroids in plants.

The morphogenic potential of nodal explants of Piper aduncum and Piper hispidinervum (Piperaceae) was investigated and a protocol for rapid micropropagation is described. An experiment based on the saline formulation of Murashige and Skoog (MS) and Wood Plant medium (WPM) combined with different N6-benzylaminopurine (BAP) and 1-naphthalene acetic acid (NAA) concentrations was evaluated. After determining the optimal concentration of growth regulators, the multiplication rates for the species, which were distributed in five subcultures, was assessed, and the number of plantlets produced in this period was recorded. After assessing the plants from all five subcultures, plantlets with well-developed root and shoot systems were transferred to pots containing substrate for acclimatization. The culture of nodal segments of P. aduncum and P. hispidinervum on hormone-free medium was shown to be a suitable method for micropropagation due to the high multiplication rate and good plant development. The use of BAP or BAP + NAA resulted to formation of vitrified multiple shoots and callus formation at the base of the microcuttings. Even at concentrations lower than 1 mg L-1 , the use of BAP resulted in vitrified multiple shoot and callus formation, without significantly improving the multiplication rates. For both species, the first subculture resulted in the greatest number of axillary buds, and mainly for P. hispidinervum, the MS medium was the most appropriate for in vitro multiplication of microcuttings. The species showed 100% root formation, and acclimatization of plants from the fifth subculture in a greenhouse resulted in 100% survival.

O presente estudo objetivou testar a sacarose e o ácido indolbutírico (AIB) no enraizamento de microestacas de teca (Tectona grandis L.f.). As bases das microestacas foram imersas nas soluções e, em seguida, plantadas nos tubetes com substrato à base de casca de pinus bioestabilizada. O experimento foi dividido em três fases: casa de vegetação, casa de sombra e pleno sol. Foram avaliados: comprimento da parte aérea (cm), diâmetro do colo (cm) e peso da matéria fresca (mg). Os resultados demonstraram haver interação entre cultivo e sacarose apenas na casa de vegetação (primeira avaliação) para as três variáveis analisadas. A concentração de AIB não apresentou diferença entre as avaliações e o uso de sacarose mostrou-se necessário principalmente quando houve sua redução no cultivo in vitro.
