Ipomoea reniformis Chaos is claimed in Indian traditional medical practice to be useful in the treatment of epilepsy and neurological disorders. In the present study, pretreatment effect of methanolic extract of Ipomoea reniformis on epilepsy and psychosis was evaluated in rodents using standard procedures. Besides evaluating epileptic and behavioral parameters, neurotransmitters such as Gamma-Amino Butyric Acid (GABA) in epilepsy and in psychosis dopamine, noradrenaline and serotonin contents in the rodent brain were estimated. The extract pretreatment reduced maximal electro shock; Isoniazid (INH) and Pentylenetetrazole (PTZ) induced seizures and also significantly inhibited the attenuation of brain GABA levels by INH and PTZ in mice. These results suggested that the observed beneficial effect in epilepsy may be by enhancing the GABAergic system. The test drug also inhibited the apomorphine induced climbing and stereotyped behavior and showed significantly reduced levels of brain dopamine, noradrenaline and serotonin which may be due to blocking of central dopaminergic, noradrenergic and serotonergic pathways or by enhancing the GABAergic system. The results obtained in present study suggest that the title plant possesses antiepileptic and antipsychotic activities in rodents.

Coenzyme Q10 is a natural antioxidant and scavenger of free radicals. In the present study, we examined the effect of coenzyme Q10 on paraoxonase 1 (PON1) activity, lipid profile, atherogenic indexes and relationship of PON 1 activity by high-density lipoprotein (HDL) and atherogenic indexes in gentamicin (GM)-induced nephrotoxicity rats. Thirty Sprague-Dawley rats were divided into three groups to receive saline; GM, 100 mg/kg/d; and GM plus coenzyme Q10 by 15 mg/kg i.p daily, respectively. After 12 days, animals were anaesthetized, blood samples were also collected before killing to measure the levels of triglyceride (TG), cholesterol (C), low-density lipoprotein (LDL), very low density lipoprotein (VLDL), HDL, atherogenic indexes and the activities of PON1 of all groups were analyzed. Data were analyzed by non-parametric Mann-Whitney test (using SPSS 13 software). Coenzyme Q10 significantly decreased TG, C, LDL, VLDL, atherogenic index, atherogenic coefficient and cardiac risk ratio. HDL level and PON1 activity were significantly increased when treated with coenzyme Q10. Also, the activity of PON 1 correlated positively with HDL and negatively with atherogenic coefficient, cardiac risk ratio 1 and cardiac risk ratio 2. This study showed that coenzyme Q10 exerts beneficial effects on PON1 activity, lipid profile, atherogenic index and correlation of PON 1 activity with HDL and atherogenic index in GM -induced nephrotoxicity rats.

Background: Oxidative stress plays a crucial role in the pathogenesis and progression of many diseases, including cardiovascular disease (CVD) and diabetes mellitus. Oxidative stress results from an imbalance between free radical formation and the protective antioxidant mechanisms. The latter mechanisms include superoxide dismutases (SODs) and glutathione peroxidases (GPx) that scavenge excessive ROS and protect cells against excess ROS production. The aim of current study was to determine the serum levels of SOD and serum GPx mRNA as well as the serum prooxidant-antioxidant balance in CVD patients. Method: A total of 103 subjects were recruited, with ≥50% stenosis (Angio + ) or <50% stenosis of one or more coronary arteries by angiography (Angio − ). The expression levels of SOD and GPx in serum were measured using real time PCR. Biochemical-analyses (e.g., triglycerides; high-density lipoprotein cholesterol; low-density lipoprotein cholesterol; fasting-blood-glucose) were determined in all the subjects. Associations of SOD and GPx levels with biochemical and anthropometric characteristics were assessed together with evaluation of the serum pro-oxidant-antioxidant balance (PAB). Results: CVD subjects had a significantly higher level of fasting blood glucose (FBG), TC, LDL-C, TG and hs-CRP levels, as compared to control subjects.
