Correlation of serum biomarkers (TSA & LSA) and epithelial dysplasia in early diagnosis of oral precancer and oral cancer

Abstract: The results suggested that serum levels of TSA and LSA progressively increases with grades of dysplasia in precancerous groups and cancer group, when compared with healthy controls. These glycoconjugates, especially LSA has the clinical utility in indicating a premalignant change.

“…Aligned with these findings, other serum tumor markers have been accepted as valuable tools for prognosis and treatment monitoring over the last two decades. Serum tumor markers such as squamous cell carcinoma antigen (SCCA) (Deng et al, 2012), carcino-embryonic antigen (CEA) (Zhang et al, 2012), and lipid associated sialic acid (LASA) (Sawhney and Kumar, 2011) have been examined for their value in detecting head and neck and other cancers. However, due to their low sensitivity, these markers are not always clinically useful for the diagnosis of OSCC (Sawant et al, 2008).…”

“…Sialic acids are also important terminal sugars in cell membrane glycoproteins and glycolipids (31). Previous studies have showed elevated levels of serum and salivary sialic acid in various carcinomas, including oral pre-cancer and OC (32)(33)(34)(35)(36)(37). Unfortunately, sialic acid was not among these 19 metabolites that we managed to identify and quantify in the present study.…”

“…Downregulation of p21 and p27 by arecoline through reactive oxygen species (ROS)/mTOR complex 1 (ROS/mTORC1) pathway may facilitate G1/S transition of the cell cycle which promotes error‐prone DNA replication . Arecoline‐induced overexpression of human telomerase reverse transcriptase (hTERT) and reduction in BRCA1 and BRCA2 tumour suppressor genes may also contribute to the malignant transformation of OSMF . Moutasim et al found upregulation of the integrin receptor αvβ 6 by arecoline in oral keratinocytes, which may strongly influence their migration and induce invasion.…”

“…Although new pathological markers are ever emerging, epithelial dysplasia has to date been the most reliable indicator for predicting potential malignant transformation in OSMF. The incidence of dysplasia in OSMF varies from 7 to 26% . A long‐term follow‐up study of 17 years reported a 7.6% malignant transformation rate with a reported range in the literature from 7 to 13% .…”
