Correlation between variable-number tandem-repeat-based genotypes and drug susceptibility in Mycobacterium avium isolates

Abstract: Little is known about the correlation between genotype and drug susceptibility in Mycobacterium avium (Mav) strains isolated from patients with Mav infections. To examine whether drug susceptibility profile of Mav is associated with genotype, we carried out variable-number tandem-repeat (VNTR) typing and drug susceptibility testing for Mav isolates from Japanese with nodular-bronchiectasis (NB)-type and cavitary disease (CA)-type diseases. We performed M. avium tandem repeat (MATR)-VNTR typing and drug suscept… Show more

“…However, 3 strains, DH-2, IH-068, and NN-127, were grouped into the clusters containing strains isolated abroad, suggesting that the strains in cluster I are genetically similar to those isolated abroad. Previous studies reported the association between genotype and drug susceptibility in NTM (29,30). In the present study, significant differences in the log 2 MICs of seven of eight of the drugs tested, with the exception being ethambutol, were observed among the isolates in the three clusters grouped by VNTR typing of M. avium subsp.…”

“…No association of a specific VNTR pattern with a particular clinical feature, such as the pulmonary or lymph node infection, was observed. Further studies will provide information about possible association between MAH genotypes and other clinical characteristics in our geographical region; about this, other studies report that in Japan M. avium genotypes were associated with disease progression or therapeutic response (Kikuchi et al, 2009;Kikuchi et al, 2014), or that there was no significant association between MAH genotypes isolated from Korean patients and clinical manifestation or treatment response (Kim et al, 2016;Kim et al, 2012;Tatano et al, 2012).…”

“…The tree was generated using the UPGMA method by the MIRU-VNTRplus web application available at www.miru-vntrplus.org. 2018) and a further study has reported a significant association between VNTR genotypes and susceptibility to quinolones (Tatano et al, 2012).…”

“…Interestingly, the presence of IS Mav6 was associated with drug resistance to MXF in this study. Tatano et al reported an association between the VNTR genotype and susceptibility to quinolones and EMB [ 40 ]. Dvorska et al found no relationship between IS 1311 and IS 1245 -based RFLP genotypes and drug susceptibility in MAC isolates [ 41 ].…”
