Correlation between mutations in p53 gene and protein expression in human lymphomas

Abstract: In these cases, the possibility that mutations occur outside the exons studied has been tested and the entire coding sequence analyzed. Only one case showed a mutation in exon 10, and we found two cases carrying a polymorphism in exon 4 and in intron 10.We conclude that mutations in p53 occur mainly in high-grade B-cell NHLs. Although not limited to a specific subtype of lymphoma, they may be rare in Hodgkin's disease and in low-grade lymphomas. The discrepancies between overexpression and presence of mutation… Show more

“…[48][49][50] Expression of p53 protein has previously been demonstrated in 13% to 70% of DLBCLs, 9,12,[50][51][52] and has been frequently demonstrated in the absence of a mutation. 49,53,54 This may alternatively be explained by p53 stabilization by mechanisms such as mdm2 interaction. [55][56][57] Wild-type p53 activity induces mdm2, 58 functioning to degrade p53 as a negative feedback control mechanism, 59 and high levels of mdm2 have been demonstrated in association with p53 expression in high-grade NHL.…”

“…The application of this analysis depends on the characteristics and specifi city of the monoclonal or polyclonal antibodies for p53. 15,16,20,26,29 Latent EBV infection was initially suggested as a possible cause for p53 mutation. This hypothesis has never been confi rmed in clinical studies.…”
