Endosulfan (ENDO) is a widely used organochlorine (OC) pesticide and persistent organo-pollutant. Epidemiological studies have shown that high levels of OC exposure were related to colorectal cancer (CRC) incidence. The objectives of the present study were to evaluate histological changes in the colon, as well as in in situ expression of b-catenin and P-selectin, and serum levels of select pro-inflammatory cytokines in mice administered ENDO; there is a relationship between increased serum IL-6 and P-selectin levels in CRC patients and aberrant b-catenin signaling is important in initiation/maintenance of most CRCs. Mice were exposed to ENDO (at dose < LD 50 ) orally once a week for up to 24 weeks, and monitored (inclusive) for a total of 42 weeks. The experiment was comprised of three groups, one that did not receive ENDO (olive oil vehicle), one administered 2 mg ENDO/kg/week and a positive control (for induction of CRC) given a weekly 20 mg 1,2-dimethylhydrazine (DMH)/kg injection. The results indicated that oral administration of ENDO provoked moderate inflammation starting at six weeks, and severe colonic inflammation with an appearance of dysplastic formations (aberrant crypts) in mice treated with ENDO (or DMH) for 12 weeks or longer. Serum IL-6 levels significantly increased starting at six weeks and rose to a peak of 15-fold higher than in controls at 42 weeks; TNFa levels likewise significantly increased, with a later peak (%four-fold higher than controls) at 30-42 weeks. Immunohistochemical analysis of the colon also showed that expression of b-catenin and P-selectin increased with length of exposure to ENDO. Taken together, the results indicate that continued repeated oral exposure to ENDO induces increased expression of b-catenin and P-selectin, inflammation in the colon, and, ultimately, local tissue dysplasia. In the past few decades, a large number of pesticides have been widely used for plant protection. Pesticides may enter non-target organisms through multiple ways and bring potential health risks. There is a dense and diverse microbial community in the intestines of mammals, which is called the gut microbiota. The gut microbiota and its metabolites play vital roles in maintaining the health of the host. Interestingly, many studies have shown that exposure to multiple pesticides could affect the gut microbiota of the host. However, the roles of gut microbiota and its related metabolites in the host health effects induced by pesticide exposure of non-target organisms need further study. We reviewed the relationships between pesticide exposure and host health effects as well as between the gut microbiota and host health effects. Importantly, we reviewed the latest research on the gut microbiota and its metabolites in the host health effects induced by pesticide exposure.

O uso intensivo de agrotóxicos no Brasil impõe o risco do aparecimento de efeitos tóxicos diversose altamente deletérios,como os efeitos sobre o sistema imunológico e endócrino. Os testes preconizados pelas diretrizes nacionais einternacionais possuem limitações para uma avaliação preditiva completa do amplo espectro de moléculas, receptores, células e órgãos-alvo dos agrotóxicos com essas propriedades. Além disso, a interação entre os sistemas nervoso, endócrino e imunológico acaba por difi cultar o estudo desses efeitos quepodem impactar a reprodução, processos metabólicos e a resistência a patógenos e o combate a tumores. A exemplo do que ocorre com medicamentos, a revisão periódica do registro de agrotóxicos no Brasildeve ser adotada em caráter urgente para identifi car e proibir o uso de agrotóxicos que sejam extremamente danosos à saúde. Apesar de prevista na lei, a revisão do registro demandada pelos órgãos da saúde e do meio ambiente passa por processo de judicialização, impedindo ou atrasando em vários anos a sua conclusão. Também se mostra urgente uma política governamental para a transição do atual modelo de produção vinculado ao agronegócio para um de produção de base agroecológica.
